오퍼란도 (operando) 분광전기화학은 전기화학적 반응성과 분자 수준의 광학적 신호를 실시간으로 측정할 수 있는 기법으로, 반응 메커니즘에 대한 직접적인 통찰을 제공한다. 본 연구에서는 비타민 E (Vit E)의 산화 과정에서 오스뮴 산화환원 매개체의 존재 하에 나타나는 전위 의존적 스펙트럼 변화를 관찰하기 위해, UV–vis 흡수 분광법과 전기화학적 제어를 결합하였다. 오스뮴 복합체는 산화 상태에 따라 구분되는 특징적인 흡수 밴드를 나타내었으며, 이를 통해 인가 전위에 따른 매개체의 거동을 정량적으로 관찰할 수 있었다. 오스뮴 복합체와 비타민 E 가 함께 존재하는 용액에서는, 매개체 단독 시스템에서 관찰되는 산화된 오스뮴 종의 축적이 현저히 억제되는 현상이 나타났다. 이러한 결과는 전극과 비타민 E 사이에서 오스뮴 매개체가 빠르게 전자를 전달하며 순환하고 있음을 의미하며, 촉매적 turnover 가 활발히 일어나고 있음을 시사한다. 이러한 오퍼란도 분광학적 신호는 오스뮴 복합체가 과전압을 낮추면서 비타민 E 의 산화를 촉진하는 EC' (전기화학– 촉매) 메커니즘이 작동함을 직접적으로 입증한다. 본 결과는 전기촉매로 작용하는 오스뮴 복합체의 반응 메커니즘을 규명할 수 있었다.

Operando spectroelectrochemistry enables real-time correlation of electrochemical reactivity with molecular-level optical signatures, providing direct insight into reaction mechanisms. Here, UV–vis absorption spectroscopy was coupled with electrochemical control to monitor potential-dependent spectral changes during the oxidation of vitamin E (Vit E) in the presence of an osmium redox mediator. The osmium complex exhibited distinct absorption features corresponding to its redox states, enabling quantitative tracking of mediator dynamics under applied potentials. In solutions containing both the osmium complex and Vit E, the expected accumulation of the oxidized osmium species was markedly suppressed compared to the mediator-only system. This behavior indicates rapid electron shuttling between the electrode and Vit E, consistent with catalytic turnover of the mediator. These operando spectral signatures provide direct evidence for an EC' (electrochemical–catalytic) mechanism in which the osmium complex promotes Vit E oxidation at reduced overpotentials. The results demonstrate the utility of spectroelectrochemistry for mechanistic identification of mediated electrocatalytic processes.
