In this study, we developed a specific cell-targeting gene delivery system with cationic polymer as non-viral vector. Artery wall binding peptide (AWBP), a specific targeting peptide, was conjugated to poly (ethylene glycol)-grafted-poly (L-lysine) (PEG-g-PLL) to enhance the transfection efficiency to artery wall cells. Characters of the physical and biochemical properties of the synthesized AWBP-PEG-PLL and AWBP-PEG-PLL/p DNA complex were analyzed through H-NMR, gel electrophoresis, atomic force microscopy (AFM) and from both bovine aorta endothelial cells (EC) and smooth muscle cells (SMC). We expected that AWBP-PEG-PLL, a gene delivery system for arterial wall targeting, will be used for genetic treatment of cardiovascular diseases. After this study, it will be continuosly investigated for the synthesis of gene carrier using low molecular weight water soluble chitosan (LMWSC).
