pCrc44-rbcL-AAD, consist of rbcL Chlamydomonas reinhardtii chloroplast DNA and aminoglycoside adenyl transferase promoter from (aadA) structural gene from E. coli in C. reinhardtii chloroplast DNA atpB region, was transformed to atpB region of C. reinhardtii chloroplast DNA by microprojectile bombardment. 37 clonies were selected as photosynthesis competent cell lines on HS minimal medium, among them 5 clonies expressed photosynthesis competent and spectinomycin resistant as a result of expression of atpB and aadA genes. All transformed cell lines showed two distinctively hybridized bands, 4.1kb and 3.1kb, by using 0.5kb atpB fragment for probe. It meant that all of them were heterokaryon of normal and aadA transformed cells. RNA gel blot analysis showed that expression of rbcL promotor was inhibited during light illumination.
