Hippocampus is known as involving the learning and memory. It is also known that hippocampus is the major brain region where the neurogenesis is occur. Hippocampus has neural progenitor cells which can differentiate to many brain cell types.
Hib5 hippocampus cell line is a immortalized by temperature sensitive T antigen and has characteristics of differentiation at 39 ℃. To analyze basic characteristics of Hib5 cells, HPLC analysis using fluorescence detector was performed. In this study, various monoamine contents (norepinephrine, dopamine, DOPAC, serotonin, tryptophan, 5-hydroxytryptophan) were analyzed.
As a results, the monoamine contents of Hib5 cells were as belows;
Tryptophan and serotonin were detected but dopamine, DOPAC, and 5-hydroxytryptophan were not detected. Norepinephrine was also not detected.
We think that these results would provide basic data on the characteristics of Hib5 hippocampus cell line and HPLC with fluorescence detection would provide us very useful tools of monoamine analysis.
