This study investigated the culture characteristics of Sabulilitoribacter sp. in a batch mode and sought to enhance zeaxanthin productivity through statistical optimization of the culture medium. First, optimal conditions for cell growth and zeaxanthin production were determined by adjusting incubation temperature and initial pH, with the best results observed at 30°C and pH 7.0. To identify suitable carbon sources for promoting zeaxanthin synthesis, batch cultures were supplemented with 30 mM of glucose, glycerol, galactose, fructose, xylose, or sucrose; however, none of these carbon sources displayed a significant improvement in growth or productivity.
For medium optimization, a fractional factorial design (FFD) was first applied to screen the eleven components of marine broth (MB), identifying peptone, yeast extract, and NaHCO₃as the main factors, influencing zeaxanthin production. These variables were then further optimized using a central composite design (CCD), resulting in predicted maximal zeaxanthin production of 3.395 mg/L with initial concentrations of peptone 8.68 g/L, yeast extract 4.068 g/L, and NaHCO₃0.253 g/L. Flask-scale validation yielded 3.594 mg/L zeaxanthin, approximately double that obtained in MB. Batch culture using the optimized medium in a 5-L fermenter further increased zeaxanthin titer to 5.423 mg/L, representing a 50% improvement over flask culture.
The effect of dissolved oxygen (DO) was subsequently evaluated in batch fermentations at 10%, 20%, and 30% pO2. Maximum zeaxanthin production was observed at 20% pO2, while higher oxygen levels resulted in decreased yields.
These results demonstrate the potential for developing a microbial fermentation process for zeaxanthin production by novel marine bacteria, Sabulilitoribacter sp..
