The immunized six calves(5-6 month old) with 100 /dose of 34 kDa, 45 kDa polypeptide and Theileria sergenti merozoite vaccine(protein contend 100 /dose) which were mixed with aluminium hydroxide to elicit antibodies were challenged in field with tick in endemic pasture of theileriosis from March to September 1994 respectively.
All animals were monitored for the erythrocyte numbers, parasitemia, hematocrit and the specific antibody reactions.
The vaccinated cattle with 34 kDa polypeptide showed more slightly changes in the erythrocyte numbers, hematocrit, hematological changes after vaccination and challenged than those of nonvaccinated cattle.
The cattle were challenged with the tick in the pasture during the summer. The immunological responses demonstrated that vaccination elicited a significant. specific antibody to T. sergenti merozoite soluble antigen due to the 34 kDa and 45 kDa polypeptides vaccine. Immunization with the 34 kDa polypeptide inhibited the production of antigen in cattle infected naturally with T. sergenti while the specific antibody still detected the antigen in the non-immunized cattle. Immunization with 45 kDa polypeptide also inhibited the production of the specific antibody induced by 34 kDa antigen from T. sergenti. However it produced 75 kDa antigen. The vaccine derived from the merozoite of T. sergenti produced the 34 kDa. It was very sensitively detected. The control group which had natural theileriosis revealed 75 kDa and 43 kDa and 32 kDa. The specific bands, 75 kDa antigen from the vaccine derived from the merozoite and 34 kDa antigen from the synthesized polypeptide vaccine were not showed in the natural infection. The vaccine derived from the synthetic polypeptide were evaluated for efficacy and imunogenicity in the pasture infected with tick.
