Objects : As classic hypnotics had been reported to have adverse effects of cognitive problems, daytime drowsiness, addiction and others, non-benzodiazepine hypnotics received much clinical attention recently and zopiclone, a cyclopyrrolone derivative, has been introduced as a new hypnotic, The purpose of this study is to evaluate the effects of zopiclone on sleep by polysomnography.
Methods : The subjects were 13 normal adult males in their twenties. They received placebo and zopiclone 7.5㎎ alternatively in a double blind cross over design and on each night, their sleep were recorded by ploysomnography. Assessments by using subjective symptoms questionnaire were performed to check the residual effects of zopiclone on the following day.
Result :
1) There was no significant difference of total sleep time between placebo and zopiclone night.
2) Sleep efficiency was increased after the administration of zopiclone(p〈0.05).
3) Sleep latency was not decreased after the administration of zopiclone.
4) Zopiclone increased deep sleep, i.e., the sum of stage 3 and 4 sleep(P〈0.01).
5) Zopiclone did not change REM sleep significantly.
6) Zopiclone reduced wake time and frequency during sleep(P〈0.01).
7) Zopiclone did not cause daytime drowsiness but produced bitter taste more than placebo (P〈0.01).
Conclusion : Zopiclone is found to have effects of maintaining sleep and increasing deep sleep and sleep efficiency, but no significant sleep-inducing effects. REM sleep does not seem to be affected significantly by zopiclone. Some of the subjects reported bitter taste after the administration of zopiclone.
