    지방조직 내 플라스말로겐 대사는 체내 대사 항상성 유지에 핵심적인 기능을 수행하며, 이 경로의 이상은 비만에 수반되는 대사 질환의 병태생리와 밀접하게 연관되어 있음이 보고되고 있다. 본 연구에서는 식이유도 비만 환경에서 고지방식이 섭취가 백색 지방조직 내 리소플라스말로겐 (lysoplasmalogen) 종의 감소를 유도하며, 동시에 리소플라스말로겐 분해효소로 추정되는 transmembrane protein 86A (TMEM86A)의 발현이 유의하게 증가함을 확인하였다. Untargeted lipidomic 분석을 통해, 지방세포 특이적 TMEM86A 결손 마우스 (TMEM86A AKO)에서는 플라스메닐 리소포스파티딜에탄올아민 18:0 (LPE P-18:0)을 포함한 다양한 리소플라스말로겐 종의 조직 내 축적이 유의하게 증가함을 확인하였다. 특히 LPE P-18:0은 포스포다이에스터레이스 3B (PDE3B) 활성을 억제하여 세포 내 cAMP 농도를 높여 PKA 경로의 활성이 증가하는 현상과 밀접하게 연관되어 있었다. TMEM86A의 결손은 미토콘드리아 산화 기능과 전신 에너지 소비를 증가시켜, 고지방식이로 유도된 대사 이상을 효과적으로 개선하였다. 나아가, LPE P-18:0의 외인성 투여는 TMEM86A 결손 모델에서 관찰된 대사적 개선 효과를 in vitro 및 in vivo 모델에서 효과적으로 재현하였다. 흥미롭게도, 비만 환자의 지방조직에서는 LPE P-18:0 수준이 유의하게 감소되어 있었으며, 이는 TMEM86A 활성을 저해하거나 리소플라스말로겐을 보충하는 전략이 비만 관련 대사질환에 대한 잠재적이 치료 접근법으로 작용할 수 있음을 시사한다.

    Plasmalogen metabolism within adipose tissue plays an essential role in maintaining metabolic homeostasis, and its disruption has been linked to obesity-related complications. In the context of dietary obesity, we observed that high-fat diet (HFD) consumption leads to a reduction in lysoplasmalogen species in white adipose tissue, accompanied by a marked upregulation of transmembrane protein 86A (TMEM86A), a putative lysoplasmalogenase. Untargeted lipidomic profiling revealed that adipocyte-specific TMEM86A-knockout (AKO) results in a significant accumulation of lysoplasmalogen contents including plasmenyl lysophosphatidylethanolamine 18:0 (LPE P-18:0) in adipose tissue. Surprisingly, this alteration in lipid composition was associated with enhanced cAMP signaling due to reduced phosphodiesterase 3B (PDE3B) activity, leading to augmented protein kinase A (PKA) pathway activation. TMEM86A deficiency also promoted mitochondrial oxidative capacity and whole-body energy expenditure, thereby mitigating the adverse metabolic effects typically induced by HFD feeding. Remarkably, administration of LPE P-18:0 recapitulated many of the metabolic benefits observed in TMEM86A-deficient models, both in vitro and in vivo. Notably, adipose tissue from individuals with obesity exhibited significantly reduced LPE P-18:0 levels, supporting the notion that inhibiting TMEM86A activity or supplementing lysoplasmalogens may represent viable strategies for counteracting obesity-associated metabolic disease.
