In June of 2009,influenza virus originated from swine virus started in Mexico was declared as pandemic. As of October 2010,more than 18,000 people were dead in 214 countries, and 750,000 people were confirmed with influenza in Korea, where 270 people were dead with 0.035% death rate. In order to study immune mechanisms, resistance patterns and prediction of influenza viruse varia仕on,we characterized virus proteins and receptors. ISED (Influenza Sequence & Epitope Database) was improved with more data of viral genes including HA, NA, NA, MP, PB2, PB1, PA, NP, and NS. It has now 54,129 genes with both A and B types. The updated genes of the 2009 A(H1N1) viruses were divided into three groups. The novel H1N1 viruses belong to swine viruses, but were found to evolve very rapidly with high variation of sequences. A/Korea/01/2009 was found to belong to group 2 and it follows A/New York/06/2009 as the second most frequent strain. We obtained the HA genes from 2009 A(H1N1) and seasonal H1N1 and H3N2, and expressed the recombinant proteins using insect cell culture system. The pandemic and seasonal HA proteins were crystallized and the structures were determined at 2.7 and 2.8 A resolutions, respectively. The HA proteins were characterized with respect to protein stability. It wasfound that seasonal HA proteins have melting temperatures 5-10 degrees higher than thatof the 2009 A(H1N1) HA protein and they were less sensitive to proteolytic cleavage than the 2009 HA protein. In order to present data on geographic maps, influenza data occurred in Korea from Nov 2004 to Jan 2010 were analysed and data correlation was quantitated to install geographic distribution and propagation models of influenza. The national and regional distribution as well as propagation models based on registered hospitals were also represented on the maps. MMR and DC-SIGN as a receptor for influenza virus were expressed in insect cell and Escherichia coli as a CRD region, respectively, and refolded to be puri典ed. MMR, its CTLD4-CTLD5 region, was expressed in insect cells and characterized to form a complex with H1N1 or H3N2 HA in the presence of calcium ion.
