한국에 자생하는 바위솔속 식물들의 유전적 근연관계를 구명하기 위하여 여러 지역에서 수집한 22종류의 종들을 사용하여 RAPD 분석을 실시하였다. 60개의 random primer와 22개의 바위솔속 수집종의 genomic DNA를 가지고 RAPD 분석을 하여 쉽게 판별이 가능하며 재현성이 있는 PCR 산물을 증폭하는 10개의 primer를 선발하였다. 10개의 primer들을 사용하여 재현성이 있는 52개의 밴드를 얻었으며, 그 중에서 다형성을 보인 밴드가 43개로 82.7%의 polymorphisms을 나타내었다. UPGMA의 분석방법을 이용하여 dendrogram을 작성한 결과 18종의 바위솔속 식물은 3개의 군(A, B, C 군)으로 집괴되었으며, 잎의 형태적 특성을 기초로 한 전통적인 분류의 결과와 일치하였다. 그러나 4종은 어떤 분류군에도 속하지 않았다.
안면도 수집종(9번)을 제외한 바위솔(3~9번)의 유사도 지수는 84.2~92.3%로 연화바위솔(18-22번; 66.3~73.9%)이나 둥근바위솔(12~17번; 69.7~83.7%)에 비해 높게 나타났다. C군에 속하는 바위솔은 형태적으로 유사하며, 공통적으로 엽형은 피침형, 엽두는 예철두이며, 1~2mm의 가시를 갖는 형태적 특성을 가지고 있다. 그러므로 바위솔은 지역종간에 변이가 적은 비교적 유전적으로 안정된 종으로 생각된다. A군인 연화바위솔(18~22번)은 유사도가 66.3~73.9%로 낮게 나타났다. A군에 속한 식물들의 잎은 예두, 둔두 혹은 원두의 엽두 형태를 가진 도란형이나 타원형으로 형태가 다양하다. 그러나 잎의 끝에 가시가 없고 회분빛녹색의 잎을 갖는 등 다른 군과 구별되는 특성을 지니고 있다. B군인 둥근바위솔(O. malacophyllus)도 유사도 지수가 69.7~83.7%로 비교적 낮게 분석되었다. 형태 특성은 엽형이 도란형이며, 엽두가 예두인 잎을 가지고 있다. 또한 가시가 없고 잎에 붉은점으로 치밀하게 구성된 테두리선을 갖는 것이 이 군에 속한 식물의 특징이다. 난쟁이바위솔(O. sikokiana)과 좀바위솔은 유사도는 68.4%로 멀게 나타났다. 특히, 난쟁이바위솔은 다른종과 구별되는 독특한 형태적 특징을 갖고 있다. 포천 수집종(11번)과 매물도 수집종인 호랑이발톱(10번)은 다른 종과의 유사도가 각각 65.7%와 63.9%로 낮게 나타났다. 이 2종은 현재 명명되지 않는 새로운 종으로 추정된다.

The genetic relationship of Korean native Orostachys species collected from various regions were analysed using random amplified polymorphic DNA (RAPD) method. A total of 60 random primers were tested on eache genomic DNA of 22 Oroatachys species to examine RAPD patterns. Ten out of sixty primers tested, which amplified easily-scorabel and reproducible PCR products, were selected for gentic analysis. A total of 52 distinct amplification fragments by the selected primers were scored, 43 (82.7%) of which showed polymorphisms between species. Eighteen Oroatachys species analysed with UPGMA were clustered into three groups, A, B and C. These clusters were consistent with the conventional taxonomy based on morphological characters of leaves mainly. However, 4 species were not clustered into any group.
The similarity coefficient value of O. japonicus(no. 3~8) except Anmyeondo collected variety(no. 9) showed higher level with 84.2~92.3% than O. iwarenge(no. 18~22; 66.3~73.9%) or O. malacophyllus (no. 12~17; 69.7~83.7%). O. japonicus(no. 3~9) in group C have similar leaf morphology and characterized by lanceolate leaves, cuspidate leaf apex and 1~2mm thorn. Therefore, O. japonicus is thought to be genetically stable, and have less regional variation. O. iwarenge species in group A (no. 18~22) showed low similarity with 66.3~73.9% according to the cluster analysis. Morphological characters within group A were diversified, such as obovate and elliptical leaves with acute, obtuse and round leaf apices: However, this group could be differentiate from other groups by no thorn and grey powdery green colored leaves. O. malacophyllus species in group B (no. 12~17) showed low similarity with 66.7~83.7% according to the cluster analysis. This group is morphologically characterized by obovate leaves and acute leaf apex. This group could be differentiate from other groups by no thorn and leaf outlines formed by densely populated red dots. O. sikokiana and O. minutus had very low similarlity coefficient of 68.4% with other species studied, and had distinguished phenotypes. different to other species. Species collected from Maemuldo (no. 10) and Pocheon(no, 11) showed less than 65.7% or 63.9% similarity coefficient respectively with other species. It is assumed that these 2 species could be the new species which were not named and classified.
The clustered groups from RAPD analysis are in agreement with the conventional taxonomy based on morphological characters. Therefore, RAPD analysis can be used for providing an alternative classification system to identify genotypes and morphological characters of Oroatachys species.
