To investigate the change of Na-pump and Ca-transport in the synaptosome of hypertensive rats, right kidney of Sprague-Dawley rat was removed and left renal artery was cliped(external diameter, 0.38mm). After 3 to 4 weeks, We prepared synaptosome from cerebrum of normotensive and hypertensive rats, and carried out the experiments of ouabain binding, and Ca-uptake.
In te experiment of ouabain binding, in hypertensive rats Kd was not changed but Bmax was decreased, compared to those in the normotensive rats.
Ca-uptake in the Na-buffer was not changed, but KCI-induced Ca-uptake was increased in the both group, but more increased in the hypertensive rat.
In the present of nifedipine, KC1-induced Ca-uptake was inhibited about 30-50% in the both group.
From these results, it is suggested that decreased number of Na-pump and increased calcium transport via calcium channel might be important in the pathogenesis of hypertension.
