    SUMMARY IN KOREAN


    본 논문은 실용적인 닭 원시생식세포 배양 시스템을 확립하고 효율적인 형질전환 닭 생산을 위한 개선된 수정란 배양 기술의 개발 및 3D8 유전자를 발현하는 형질전환 닭과 비형질전환 닭 간의 생물학적 특성을 비교하는 것에 중점을 두고 수행되었다.


    제1장에서는 가금 산업의 동향과 생명공학 기술이 접목된 가금 연구 분야의 현황을 살피고, 형질전환 닭을 생산하기 위한 기술적 기반의 개요를 다루었다. 특히 CRISPR/Cas9 시스템 등 최신 유전공학 기법의 도입으로 인해 닭에서도 정밀한 유전자 편집의 가능성이 열렸다. 본 연구의 목적은 형질전환 닭 생산 과정의 실용적인 기법을 향상하고 결과적으로 태어난 형질전환 닭의 생물학적 안전성을 비교 분석하는데 있다.


    제2장에서는 형질전환 닭 기술의 발전 과정을 문헌 고찰을 통해 정리하였다. 초기 바이러스 벡터 기반 기술의 한계점과 더불어, 보다 안정적인 유전형질 전달이 가능한 원시생식세포 기반 유전자 도입법의 등장과 발전을 다루었다. CRISPR/Cas9을 활용한 유전자 편집 기술의 연구 사례도 함께 검토하였다. 또한, 닭 수정란 대리난각 배양시스템 등 수정란을 인공적으로 배양할 수 있는 기법의 장단점을 비교하였으며, 생물반응기, 질병 저항성, 면역 및 발달 모델 닭 등 다양한 형질전환 닭을 소개하였다.

    본 논문은 실용적인 닭 원시생식세포 배양 시스템을 확립하고 효율적인 형질전환 닭 생산을 위한 개선된 수정란 배양 기술의 개발 및 3D8 유전자를 발현하는 형질전환 닭과 비형질전환 닭 간의 생물학적 특성을 비교하는 것에 중점을 두고 수행되었다.

    SUMMARY


    This dissertation focuses on establishing practical chicken primordial germ cells (PGCs) culture systems, improved surrogate eggshell incubation of chicken fertile eggs for efficiently producing transgenic (TG) chickens, and understanding the biological characteristics of TG chickens expressing 3D8 single chain variable fragment (scFv) gene in comparison to non-TG chickens. The structure progresses from general introduction (Chapter 1) and literature review (Chapter 2) to experimental approaches in PGCs isolation and cultivation (Chapter 3), surrogate eggshell incubation (Chapter 4), biological evaluation of male 3D8 TG offspring (Chapter 5), and general discussion (Chapter 6).


    In Chapter 1, the global expansion of the poultry industry and its growing integration with biotechnology were discussed. The chapter outlined current approaches for generating TG chickens, including viral- and primordial germ cell (PGC)-based methods, and highlighted ongoing challenges such as limited efficiency, embryo viability, and stable PGC culture. With the advent of the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) system, new opportunities have emerged for precise genome editing in chickens. This chapter also presented the rationale and objectives of the dissertation, which aims to improve the biological safety, technical reliability, and practical feasibility of TG chicken production systems.


    In Chapter 2, a comprehensive literature review was conducted to trace the development of TG chicken technologies. The chapter covered early viral vector-mediated methods and their limitations in germline transmission, as well as the emergence of PGC-based approaches that enable more consistent and heritable gene integration. It also examined the integration of CRISPR/Cas9 systems into both viral- and PGC-mediated platforms. Various TG chicken models were reviewed, including bioreactor lines for pharmaceutical proteins
    production, disease-resistant strains, and experimental models for immunology and developmental biology. Chicken whole embryo culture systems, windowed, surrogate, and shell-less, were compared in terms of accessibility, hatchability, and experimental feasibility. The chapter provided a broad overview of current methodologies and future directions in avian biotechnology.


    In Chapter 3, a practical and streamlined approach for the stable isolation and cultivation of chicken gonadal PGCs was investigated. Establishing chicken PGCs is critical for producing genetically modified (GM) chickens, but these techniques are often time-consuming, and require labor-intensive protocols. Embryonic gonads were surgically removed, and dissociated using trypsin. Chicken gonadal PGCs were isolated by exploiting differential adhesion properties. The PGCs were maintained with mitotically inactivated SIM mouse embryo-derived thioguanine-resistant (STO) feeder cells under stable PGC culture conditions. The investigation revealed that the PGCs were positive to germline- and pluripotency-related genes, and their migration to recipients was identified through fluorescent analyses with green fluorescent dye and protein, and through polymerase chain reaction. The effectiveness of the method contributes to progress in the production of germline chimeric and GM chickens.


    In Chapter 4, the surrogate eggshell incubation system was improved by introducing a new method that utilizes the second surrogate eggs from the same poultry flock producing donor fertile eggs. Although the existing surrogate eggshell technique can support high hatching rates, it requires surrogate eggs that are approximately 30–40 g heavier than the donor fertile eggs, which limits the availability of size-qualified eggs. Moreover, externally sourced surrogate eggs may pose a risk of transmitting flock-specific or vertically transmitted avian diseases. The newly proposed method uses surrogate eggs that are only 10 g heavier than the donor eggs within the same flock. This approach resulted in a 90.7% embryo survival rate at embryonic day 4 and a 70.0% hatching rate. These results demonstrate that the method is effective for chicken whole embryo culture
    and may be applicable to the production of TG chickens.


    In Chapter 5, biological parameters collected from comparative analyses between non-TG and TG male White Leghorn (WL) offspring was investigated. The TG chickens expressing 3D8 scFv gene were used. The investigation revealed that serum alanine aminotransferase, insulin-like growth factor-1, interferon gamma, and interleukin-4 was significantly higher in non-TG male groups, but no significant differences in survival or growth were observed. These findings align with previous study conducted on non-TG and TG female WL offspring. In conclusion, this study provides foundational biological data on 3D8 scFv-expressing TG chickens, which may serve as a reference for future studies evaluating the biological characteristics of transgene expression in chickens.

    This dissertation focuses on establishing practical chicken primordial germ cells (PGCs) culture systems, improved surrogate eggshell incubation of chicken fertile eggs for efficiently producing transgenic (TG) chickens, and understanding the biological characteristics of TG chickens expressing 3D8 single chain variable fragment (scFv) gene in comparison to non-TG chickens.
