    꿀벌(Apis mellifera)은 전 세계 생태계 및 농업 생산성 유지에 핵심적인 역할을 담당하는 중요한 가축이다. 그러나 봉군붕괴현상(colony collapse disorder, CCD)의 발생으로 인해 전 세계적으로 꿀벌 개체수가 급격히 감소하고 있다. 한국에서도 2021년 겨울부터 이러한 꿀벌 개체 수 감소가 두드러지게 나타났으며, 이는 이상 기후, 농약 노출, 밀원식물 감소, 병원체 감염 등 다양한 요인들로 인해 발생하는 것으로 여겨진다. 이 중에서도 꿀벌 질병이 주요한 기여 요인으로 부각되고 있으나, 국내 꿀벌 병원체에 대한 전국적인 역학 자료와 현장에서 적용 가능한 분자 진단 기술의 개발과 검증이 부족한 실정이다.
    이에 본 연구는 국내 주요 꿀벌 병원체의 전국적 유병률을 조사하고, 가축전염병 예방법상 가축전염병으로 지정된 3종의 꿀벌 질병에 대한 현장형 원스텝 분자 진단 기법을 개발하고 검증하였다. 2020년부터 2023년까지 전국 1,378개 양봉장에서 비정상적인 임상증상을 보이는 3,707개 봉군을 대상으로 qPCR 및 RT-qPCR 분석으로 주요 꿀벌 병원체 12종의 감염 여부를 조사하였다. 그 결과, black queen cell virus (BQCV)가 2023년 기준 83.3%로 가장 높은 감염률을 보였으며, 계절에 관계없이 연중 높은 유병률을 유지하였다. deformed wing virus (DWV, 48.7%)와 Israeli acute paralysis virus (IAPV, 41.3%)는 겨울철에 특히 높은 감염률을 보였으며, CCD의 주요 특징 중 하나인 월동 실패와의 연관성을 시사하였다. 또한, 병원체의 복합감염 양상은 2020년 48.3%에서 2023년 82.3%로 뚜렷한 증가 추세를 보이며 국내 꿀벌 병원체의 감염 패턴 변화를 확인하였다. 반면, acute bee paralysis virus (ABPV)와 Kashmir bee virus (KBV)는 거의 검출되지 않았으며, 지역 간 유병률의 차이는 통계적으로 유의하지 않았다.
    확인된 병원체 유병률 양상과 현장 진단 기술의 필요성에 대응하여, 두 가지 현장형 원스텝 분자 진단 기법을 개발하였다. 첫 번째 기법은 제2종 가축전염병인 낭충봉아부패병 (SBV)을 진단하기 위한 RT-qPCR 현장 진단 플랫폼으로, 핵산 추출, 증폭, 검출 과정을 하나의 일체형 키트에 통합하였다. 성능 평가 결과, 실험실 기반 RT-qPCR과 동등한 검출 한계를 보이면서도, 낮은 농도에서 더 높은 민감도를 나타냈다. 11종 주요 꿀벌 병원체에 대한 교차 반응을 보이지 않았으며, 임상 민감도 98.4%, 특이도는 97.0%로 높은 진단 정확도를 확보하였다.
    두 번째 기법은 제3종 가축전염병인 미국부저병(AFB)과 유럽부저병(EFB)을 동시에 진단하기 위한 다중 qPCR 현장 진단 기법으로, 미국부저병의 병원체인 Paenibacillus larvae와 유럽부저병의 병원체인 Melissococcus plutonius를 각각 93.3% 및 95.2%의 민감도로 검출하였으며, 두 병원체 모두 97.0%의 특이도를 보였다. 실험실 기반 qPCR과 유사한 검출 한계를 보였으며, 임계값(Cycle threshold, Ct)이 낮아 저농도의 병원체도 안정적으로 검출할 수 있었다.
    종합적으로 본 연구는 국내 꿀벌 병원체의 감염 양상에 대한 역학적 정보를 제공하고, 낭충봉아부패병 및 부저병에 대한 현장 중심 분자진단 기법을 개발하여 실용성과 정확성을 입증하였다. 이를 통하여 꿀벌 질병 발생 경향을 파악하고 양봉장에서의 조기 진단을 가능하게 함으로써 효과적인 질병 예방 시스템과 관리 전략 수립에 기여할 것으로 기대된다. 궁극적으로 본 연구의 결과는 지속 가능한 양봉 산업과 꿀벌 질병 예방을 위한 기초 자료로 활용될 수 있을 것으로 여겨진다.

    Honeybees (Apis mellifera) play an indispensable role in global ecosystems and agricultural productivity. However, global honeybee populations have been declining significantly due to a phenomenon known as colony collapse disorder (CCD). Since the winter of 2021, South Korea has also faced a severe honeybee health crisis, attributed to multiple factors such as abnormal climate, pesticide exposure, reduced nectar and pollen sources, and pathogenic infections. Among these, infectious diseases have emerged as major contributing factors. Despite their significance, nationwide epidemiological data on honeybee pathogens and field-applicable diagnostic tools have remained limited.
    To address these challenges, this study aimed to characterize the nationwide prevalence of major honeybee pathogens and to develop field-deployable molecular diagnostic assays for three honeybee diseases designated as contagious animal diseases under the Act on the Prevention of Contagious Animal Diseases in South Korea. From 2020 to 2023, a total of 3,707 honeybee colonies exhibiting abnormal behavior were sampled from 1,378 apiaries and tested using quantitative real-time PCR (qPCR) and reverse-transcription quantitative real-time PCR (RT-qPCR) assays targeting 12 major pathogens. Black queen cell virus (BQCV) showed the highest infection rate, with 83.3% in 2023 and consistently high detection year-round. Deformed wing virus (DWV, 48.7%) and Israeli acute paralysis virus (IAPV, 41.3%) displayed elevated prevalence in winter, suggesting a link with overwintering failure, one of the key features of CCD. In addition, the pattern of co-infections showed a marked increase from 48.3% in 2020 to 82.3% in 2023. In contrast, acute bee paralysis virus (ABPV) and Kashmir bee virus (KBV) were rarely detected, and there were no significant regional differences in pathogen prevalence, likely due to South Korea's widespread migratory beekeeping practices.
    In response to the identified pathogen prevalence patterns and the need for practical field diagnostics, two point-of-care (POC) molecular diagnostic platforms were developed. The first, an RT-qPCR assay optimized for sacbrood virus (SBV)—a contagious animal disease Type II—was integrated into a portable platform (XQ SBV Dx Kit) capable of nucleic acid extraction, amplification, and detection in a single unit. This assay demonstrated laboratory-equivalent analytical sensitivity and superior detection at low viral concentrations, with no cross-reactivity against 11 other pathogens. It achieved 98.4% clinical sensitivity and 97.0% specificity.
    The second platform was a multiplex POC qPCR assay developed for the simultaneous detection of American foulbrood (AFB; caused by Paenibacillus larvae) and European foulbrood (EFB; caused by Melissococcus plutonius), both classified as contagious animal diseases Type III. The assay detected P. larvae and M. plutonius with sensitivities of 93.3% and 95.2%, and specificity of 97.0% for both. The limit of detection (LOD) was comparable to laboratory qPCR, with lower cycle threshold (Ct) values enabling robust identification even at low pathogen loads. These diseases have been prioritized for diagnostic development based on their legal designation as contagious animal diseases in South Korea and their epidemiological significance, including high mortality, rapid spread potential, and biosecurity-related movement restrictions.
    Taken together, this study provides valuable insights into the epidemiological dynamics of honeybee pathogens in South Korea and demonstrates the practical feasibility of POC molecular diagnostics for real-time use with one-step protocol in apiaries. By enabling early and accurate diagnosis, these tools support evidence-based disease control and may reduce the misuse of antibiotics and other treatments, including unregulated or inappropriate drug use in apiaries. Ultimately, this research contributes to the national preparedness for honeybee disease outbreaks and offers a scalable model for integrating surveillance and diagnostics to improve apicultural sustainability and pollinator health in the context of ongoing environmental challenges.
