A Coloring and spectrophotometric method using tetrathiocyanatocobaltate(Ⅱ) ion as a complexation reagent was developed for the identification of Coptidis Rhizoma and Phellodendri Cortex in herb medicine.
Identification tests could be carried out coloring in test tube and ultraviolet-visible spectrophotometry.
Coptidis Rhizoma and Phellodendri Cortex: with a solution of 1,2-dichloroethane in thst tube - yellowish green.
The ultraviolet - visible spectrum of base components in 1,2-dichloroethane exgibits maxima at 270, 340, 442 and 624nm and was developed for the determination of berberine or palmatine in Coptidis Rhizoma and Phellodendri Cortex.
Optimum conditions for good determination was determined: Maximum wavelength 442nm; Determination of berberine was possible to the range of 0.01mg/ml-0.10mg/ml in 1,2-dichloroethane. Calibration curve showed that the coefficient of variation was 0.999% for berberine. And determination of palmatine was possible to the range of 0.10mg/ml - 0.60mg/ml in 1,dichloroethane.
The method was rapid, accurate. It was considered that spectrophotometry could be greatly applied to the analysis of base components in Coptidis Rhizoma and Phellodendri Cortex containing relatively small amount of gerb medicine extract.
