To compare the effect of LHRH and progresterone on the secretion of hCG, 8-week and term placental explants were maintained in culture for 6 days with daily changes of media The levels of β-hCG in culture media without or with LHRH or progesterone treatment were measured by radioimmunoassay The results were as follows 1 In control group of 8-week placental explants, the sopntaneous secretion of β-hCG began to increase from the 1st day of culture and reached the highest level on the 4th day of culture, thereafter the levels gradually declined But in control group of term placental explants, the level of β-hCG continued to decrease till the 4th day of culture and slightly increased thereafter 2 The effect of LHRH on the secretion of β-HCG from 8-week placental explants was not observed significantly throughout the incubation period except on the 6th day of culture in 3200μg/ml LHRH-treated group But in term placental explants, β-hCG levels significantly increased from the 4th day of culture with dose-response manner (R=0 93, P<0 01) 3 In the effect of progesterone on the secretion of β-hCG from 8-week placental explants, β-hCG levels significantly decreased with dose-response manner (r=0 97, p<0 01) from the 3rd day of culture with 20μg/ml of progesterone treatment and on the last day of culture with 5μg/ml of progesterone treatment But no significant changes in β-hCG levels were observed throughout the entire incubation period in therm placental explants 4 Therefore, it is concluded that LHRH stimulated the secretion of β-hCG more significantly in term placental explants than 8-week placental explants, and progesterone suppressed the secretion of β-hCG more significantly in 8-week placental explants than in therm placental explants
