      Intervertebral disc is comprised of two distinct region anulus fibrosus and nucleus pulposus. Nucleus pulposus is composed of hyaline cartilage that has load-bearing strength and contains proteoglycans and type II collagens. Anulus fibrosus is composed of fibrous cartilage that bears a tension and give flexibility to the vertebrate, and it shows high expression of type I collagen. It was found that type I collagen was expressed in anulus fibrosus dominantly, aggrecan was expressed in nucleus pulposus, and type II collagen and type VI collagen were expressed over the whole intervertebral disc. Fibrotic cartilage such as AF was enhanced collagen birefringence by Picro-sirius red staining under polarized light microscopy, but hyaline cartilage such as NP was not. In this study, we suggests that the analysis of collagen fibril by Picro-sirius red staining is good method to estimate the completion level of fabricated disc.
