Loss-of-function mutations in the filaggrin gene have been clearly associated with the severity of atopic dermatitis and allergic phenotypes. Complex interactions have been described between immune abnormalities characterizing AD such as elevated Th2 cytokine levels and expression of genes involved in epidermal barrier function including filaggrin. Although excessive expression of IL-13/IL-4 is damaging to skin barrier, these cytokines may have important roles in the regulation of skin barrier function at constitutive levels of expression.
