The anti-epidermoid cancer effect of Eugenol: Targeting OR7A17 induced cell cycle arrest and apoptosis

Background: Olfactory Receptors (ORs) are increasingly recognized to be ectopically expression in non-olfactory tissues, including the skin, where they are implicated in various cellular events. Since OR7A17, known to be involved in keratinocyte biology, is an orphan receptor, the purpose of this study was to identify OR7A17 ligands and investigate their potential application in the treatment of epidermal cancer.

Methods: To investigate the OR7A17 antagonism and anticancer activity of eugenol, various tools were introduced, including virtual screening and AMDock analysis, fluo-4 Ca²⁺ influx assay, and CETSA (cellular thermal shift assay), CellTiter-Glo, EdU incorporation, colony formation, and cell scratch assay. Western blot analysis and real-time PCR analysis were performed to elucidate the molecular mechanism of eugenol.

Results: The virtual screening showed that eugenol, a natural phenolic compound derived from clove oil, occupies the binding pocket of OR7A17 receptor with a high affinity (-5.7 kcal/mol). The direct binding between eugenol and OR7A17 was verified by CETSA, demonstrating eugenol as a ligand of OR7A17 receptor. In addition, eugenol significantly suppressed intracellular Ca²⁺ influx and cell proliferation in OR7A17-overexpressing HaCaT cells. Western blot also showed that eugenol reduces OR7A17-mediated proliferation by suppressing the phosphorylation of MAPK components. Furthermore, the eugenol effects were attenuated by the knockdown of OR7A17 gene. These results indicate that eugenol acts as an antagonistic ligand of OR7A17. In an experiment comparing the expression levels of OR7A17 in A-431 cells, an epidermal cancer cell line, and HaCaT cells, a normal keratinocyte cell line, A-431 cells were found to express more OR7A17 than HaCaT cells. In addition, eugenol inhibited cell proliferation and cell migration more potently in A-431 than in HaCaT. These results suggest that A-431 cells are more sensitive to eugenol than HaCaT cells because of higher expression of OR7A17 in A-431 cells. Furthermore, treatment with eugenol induced apoptosis in A-431 cells, as evidenced by an increase in the pro-apoptotic protein Bax and a decrease in the anti-apoptotic protein Bcl-2.

Conclusion: This study demonstrates eugenol as an OR7A17 antagonist and its anti-epidermoid cancer effect is mediated by OR7A17 antagonism. These findings also suggest OR7A17 as a novel molecular target for the development of skin cancer therapeutics.
