Alginate lyase from Chlorostma xanthostigma was stabilized in vitro by optimizing buffer ingradients. The extraction buffer consists of 25mM acetate, pH 5.5, 20mM MgCl_2, 100mM NaCl and 3mM PMSF. The alginate lyase extracted with this buffer gave a decay rate of less then 1% per hr at 30℃ for at least 6h. In vitro instabihty of alginate lyase may be due to proteolytic degradation.
