      We developed tyndallized (heat-treated) lactic acid bacteria with anti-allergic effect. Tyndallization process was established with Lactobacillus rhamnosus IDCC 3201 isolated from Korean breast milk-fed infant. Thermal death times for Lactobacillus rhamnosus IDCC 3201 were evaluated to identify optimal tyndallization temperature. Then, validation of direct counting method for calculating the number of dead bacterial cells was performed. The cell counting method was successfully validated on linearity, precision, and accuracy. To evaluate immunomodulatory effect of Lactobacillus rhamnosus IDCC 3201 tyndallizate (RHT3201), cytokine production was carried out with determination of interleukin (IL) 4, IL-12, and IFN-γ in mouse splenocytes from OVA-sensitized BALB/c mice. Treatment of RHT3201 down-regulated type 2 helper T (Th2) cytokine IL-4 and upregulated type 1 helper T (Th1) cytokines such as IL-12 and IFN- γ dose dependently. The immunomodulation was induced by improving the balance between Th1 and Th2 cytokines. Therefore, we finally developed tyndallized lactic acid bacteria with superior anti-allergic effect.
