본 연구는 서식환경이 다른 참복 Takifugu chinensis, 국매리복 T. vermicularis 그리고 황복 T. obscurus 신장의 미세해부학적 특징을 비교하고자 하였다. 비뇨계는 “Y”형으로 신장, 수뇨관, 후주정맥, 방광으로 구성되었다. 신장은 좌우 한 쌍의 둥근 삼각형 형태로 “Y”형 비뇨계의 가지 끝부분에 위치하였으며, 돌출된 상단부의 두신부와 아래쪽의 체신부로 구분되었다. 두신부와 체신부는 결합조직에 의해 구분되며, 두신부는 전체 신장의 약 1/5을 차지하였다. 수뇨관과 후주정맥은 접합된 관 구조로 이들 상부의 약 1/10은 체신부에 함몰되어 있으며, 한 쌍의 체신부 말단부에서 이어져 오다가 양쪽의 수뇨관과 후주정맥이 하나로 합쳐지는 형태였다. 방광은 수뇨관의 말단부에 주머니 형태로 존재하였다. 이러한 비뇨계의 형태는 기존에 보고된 경골어류와 차이를 보였다. 네프론의 조직학적 구조는 기존에 보고된 경골어류와 유사하였으며, 신소체는 보우먼주머니가 사구체를 감싼 형태였다. 사구체는 모세혈관이 뭉친 환 형태로 모세혈관, 내피세포, 사구체 간질, 유족세포로 구성되었다. 보우먼주머니는 이중벽 구조로 내층의 편평형 상피세포와 외층의 결합조직성 막으로 구분되었다. 세뇨관은 단층의 섬모원주형 상피세포로 구성되며, 내강의 자유면에는 선조연이 발달했다. 두신부에서는 특징적으로 간신세포와 크롬친화세포가 군집된 구조로 분포하였다. 크로마핀 반응에서 간신세포는 반응하지 않아 H-E 이중염색에서 호산성을 나타냈고, 크롬친화세포는 H-E 이중염색에서 갈색으로 산화 반응하였다. 참복, 국매리복, 황복 신장의 조직학적 구성은 동일하나 사구체 분포율은 서식환경에 따라 다르게 나타났다. 신장에서 사구체 분포율은 참복 (0.13%), 국매리복 (0.16%), 황복 (0.21%) 순으로 높았으며, 광염성이 높을수록 높은 경향을 보였다. 본 연구 결과, 참복, 국매리복, 황복 모두 두신부에서 네프론을 가지며, 황복과 같은 광염성이 높은 종에서 사구체가 발달하여 염분 농도 변화에 적응하기 위한 기작으로 판단되었다.

This study aimed to compare the microanatomical characteristics of the kidneys of three pufferfish species with different habitats: Takifugu chinensis, T. vermicularis, and T. obscurus. The urinary system exhibited a distinct Y-shaped structure composed of the kidney, ureter, posterior cardinal vein, and urinary bladder. The paired kidneys were round-triangular in shape and located at the distal ends of the branches of the Y-shaped urinary system. Each kidney was divided into an anterior head kidney and a posterior body kidney, with the head kidney accounting for approximately 1/5 of the total kidney volume and being distinguished from the body kidney by connective tissue. The ureter and posterior cardinal vein formed an adjacent tubular structure, of which approximately 1/10 of the upper portion was embedded in the body kidney. This structure extended from the terminal ends of the paired body kidneys, where the bilateral ureter and posterior cardinal veins merged into a single unit. The urinary bladder was present as a sac-like structure at the distal end of the ureter. The overall morphology of the urinary system differed from that previously reported in other teleost fishes. The histological structure of the nephron was generally similar to that of teleosts described in previous studies. The renal corpuscle consisted of a glomerulus surrounded by Bowman’s capsule. The glomerulus was composed of a network of capillaries, endothelial cells, mesangial tissue, and podocytes. Bowman’s capsule exhibited a double-layered structure, consisting of an inner layer of flattened epithelial cells and an outer connective tissue membrane. Renal tubules were lined with a single layer of ciliated columnar epithelial cells, and a well-developed brush border was observed on the luminal surface. Notably, clusters of interrenal cells and chromaffin cells were characteristically distributed in the head kidney. In the chromaffin reaction, interrenal cells showed no reaction and appeared eosinophilic in hematoxylin–eosin (H–E) staining, whereas chromaffin cells exhibited an oxidative reaction and appeared brown in H–E staining. Although the histological composition of the kidney was similar among T. chinensis, T. vermicularis, and T. obscurus, the glomerular area ratio differed according to habitat. The glomerular area ratio increased in the order of T. chinensis (0.13%), T. vermicularis (0.16%), and T. obscurus (0.21%), showing a tendency to be higher in species with greater euryhalinity. These results indicate that all three species possess nephrons in the head kidney, and that species with higher euryhalinity, such as T. obscurus, exhibit more developed glomeruli, which is considered an adaptive mechanism for coping with fluctuations in salinity.
