고요산혈증(hyperuricemia)은 퓨린 대사 이상으로 인해 체내에 요산(uric acid)이 과도하게 축적되어 발생하는 대사성 질환으로, 통풍(gout), 신장질환, 심혈관 질환 등 다양한 만성 질환과 밀접하게 관련되어 있다. 이를 치료하기 위한 접근법 중 하나로 유리케이스(UOX)를 활용한 uricase enzyme therapy 가 주목받고 있으나, 해당 효소가 요산을 분해하는 과정에서 과산화수소(H₂O₂)라는 독성 부산물이 함께 생성되며, 이로 인한 활성산소종(ROS)-매개 세포 독성 및 조직 손상이 치료의 새로운 병리적 부작용으로 보고되고 있다. H₂O₂는 세포 내 산화 스트레스를 유도하여 DNA 손상, 세포 사멸(apoptosis), 염증 반응 및 조직 섬유화 등을 초래할 수 있으며, 이러한 문제는 uricase 치료의 주요한 임상적 한계로 대두되고 있다. 이를 해결하기 위해 본 연구에서는 요산 분해 효소인 UOX 와 과산화수소 제거 효소인 카탈레이스(CAT)를 자발적으로 결합시키는 새로운 모듈형 효소 복합체 전략을 제안하였다. 기존의 직접 융합(fusion) 또는 나노입자 캡슐화(encapsulation) 방식이 구조적 유연성 저하, 낮은 효소 활성, 재현성 부족 등의 기술적 한계를 지닌 반면, 본 연구에서는 생리적 조건에서도 선택적이고 안정적인 상호작용이 가능한 coiled-coil 기반 류신지퍼 모티프를 각각의 효소에 부착하여, 비융합 방식의 가역적이고 자발적인 복합체 형성을 유도하였다. 이 과정에서 결합력 강화를 위해 시스테인 변이를 도입하였다. 형성된 복합체는 Size-exclusion chromatography(SEC) 및 Dynamic Light Scattering(DLS)을 통해 그 물리화학적 특성이 입증되었으며, 기능적 분석에서도 낮은 효소 농도에서도 uric acid 분해와 H₂O₂ 제거가 기존 단일 효소 처리 대비 현저히 향상되는 결과를 보였다. 특히 H₂O₂ 제거에 있어 거의 완전한 제거 효과를 나타냈다. 이어진 HCT116 세포 기반 실험에서는 모듈형 복합체가 H₂O₂ 유발 세포 독성을 효과적으로 억제하고, 세포 생존율 증가 및 apoptosis 관련 지표(TUNEL, PI 염색)의 유의한 감소를 유도하였다. 이러한 결과는 기존 uricase 치료의 한계로 지적되던 산화 스트레스 문제를 해결할 수 있는 효과적인 전략을 제시함과 동시에, 각 효소의 고유 활성을 유지하며 복합 cascade 반응을 구현할 수 있는 플랫폼 기술로서의 가능성을 입증한다. 본 시스템은 향후 고요산혈증뿐 아니라, 다중 효소 반응 기반의 치료제 및 바이오산업 전반에 활용될 수 있는 범용적 응용 잠재력을 갖는다. 도메인 특이적 교차결합에 의한 요산분해효소–카탈라아제 융합체의 공학적 설계: 고요산혈증 치료의 새로운 전략 박 종 하 국립 부경대학교 대학원 화학융합공학부 화학공학전공

Hyperuricemia is a metabolic disorder caused by abnormal purine metabolism, leading to excessive accumulation of uric acid in the body. It is closely associated with chronic conditions such as gout, kidney disease, and cardiovascular disorders. Among the therapeutic approaches, uricase-based enzyme therapy has gained attention for its ability to degrade uric acid. However, the enzymatic reaction also produces hydrogen peroxide (H₂O₂) as a byproduct, which acts as a reactive oxygen species (ROS) and induces cytotoxicity and tissue damage. This oxidative stress has emerged as a critical unmet need in current uricase therapy. To address this limitation, we developed a modular enzyme complex strategy that enables spontaneous assembly of uricase (UOX) and catalase (CAT), an H₂O₂-degrading enzyme, without direct fusion. While conventional fusion or nanoparticle-encapsulation methods often suffer from structural rigidity, reduced enzyme activity, and poor reproducibility, our approach utilizes coiled-coil leucine zipper motifs attached independently to each enzyme to mediate selective, reversible interactions under physiological conditions. Furthermore, cysteine mutations were introduced to enhance the binding affinity between the motifs. The formation of the UOX–CAT complex was confirmed through biophysical characterization using size-exclusion chromatography (SEC) and dynamic light scattering (DLS). Functional analysis revealed that the coiled-coil- mediated enzyme complex significantly improved both uric acid degradation and H₂O₂ elimination even at low enzyme concentrations, with nearly complete removal of H₂O₂ achieved. This platform technology holds potential not only for the treatment of hyperuricemia but also for broader applications in multi- enzyme cascade systems in therapeutic and industrial contexts.
