The Study on the Effects of Cortisone to the Thiamine deficient Albino Rats
Eijkman first produced polyneuritis in 1897 by feeding chicken with white rice. Since then it has been studied by many workers and thiamine was discovered which is the main factor for protection of polyneuritis by Jansen and Donath in 1926 formular was cleared by Windaus in 1931, and at last Williams Synthesized it in 1935
Physiological action of thiamine is the activation of the enzymes of Pyruvate and α -ketoglutarate decarboxylation and it has been known that thiamine pyrophosphate is active form of thiamine
The purpose of this experiment is to compare the tissue respiration with succinic dehydrogenase activities in the livers of thiamine deficient rats with and without administration of cortisone and to investigate the fact as, cortisone on the thiamine deficient rats
Rats were grouped in four as follows;
I. Control group.
II. Cortisone administered control group.
III. thiamine deficient group.
IV. Cortisone administered, thiamine deficient group.
After the rats were fed for 3 weeks, all rats were sacrificed and O₂ consumption and succinic dehydrogenase activities of the livers were determined and the Vitamin C level in blood, adrenal gland and liver was determined and the results were as follows.
1) The growth of the thiamine deficient rats was poor compared with that of the control and the growth of the cortisone administered thiamine deficient rats was liver poorer than that of the thiamine deficient rats which were not administered cortisone but the cortisone administered control rats which were not deficient of thiamine, grew almost well as the control did.
2) The weight of the livers of the thiamine deficient rats reduced compared with that of control. And the administration of cortisone reduced the weight the more
3) The Vitamin C levels in blood and liver of the thiamine deficient rats were lower than those of the control, and cortisone reduced the Vitamin C levels the more
4) There were no differencies between O₂ consumption and succinic dehydrogenase activities of the livers of thiamine deficient rats with and without administration of cortisone and control groups with and without administration of cortisone
From this fact, it may be assumed that thiamine deficiency and cortisone did not change much at the glucose oxidation in this experiment.
