Abstract
Bivalves such as oyster have sessile property. So they can expose easily to pollutants, stress, and organism. They can cause oxidative stress.
Free radicals cause oxidative stress and lead to cellular degeneration. There is generally the balance between free radicals and antioxidants. But if free radicals are rapidly produced, the balance between both breaks by the relative lack of antioxidants. White muscle disease(WMD) is known as a disease caused by oxidative stress in higher vertebrates. Vitamin E deficiency is fundamental cause in WMD. This study is aimed to observe tissue response in the oyster after CCl₄ injection which evokes oxidative damage. This study was performed to know the histopathological effect of CCl₄ in the cultured Pacific oyster(Crassostrea gigas). 10% CCl₄ diluted olive oil was injected into the body. PBS and olive oil were used in negative and positive controls respectively. Sampling of 10 individuals was done just before injection, 12 hours, 24 hours, 72 hours, and 120 hours after from each experimental groups. After the injection of CCl₄, a number of hemocytes were accumulated in the heart with the degeneration of muscle. Hemocyte infilteration index (HII) was significantly the highest 24 hours after the injection. After 24 hours, the degree of accumulation tended to be gradually recovered toward normal. Other organs and tissues showed no marked change in the number of hemocytes and other histological structures. There were no histopathological alteration in both control groups. In this study the oxidative damage artificially induced by CCl₄ injection was confined to the heart muscle in Pacific oyster. Therefore it was suggested that histological findings of the heart muscle of Pacific oyster could be used for monitoring of oyster health under the oxidative environment.
