    Ⅰ. 연구개발의 목적 및 필요성
    Amyotrophic Lateral Sclerosis (ALS: 근위축성측삭경화증)는 운동신경세포가 파괴됨으로써 근육들이 운동신경의 자극에 대해 적절한 운동기능을 나타내지 못하게 되는 퇴행성 신경질환의 하나임. 본 연구는 ALS의 응집체의 기능과 형성과정의 이해를 통해 발병기전을 연구하고 돌연변이 SOD1과 상호 결합하는 단백질과 SOD1의 결합부위를 동정하고 동정된 단백질의 절편을 사용하여 세포수준과 ALS 동물 모델에서 경쟁적 저해제로서 ALS치료제로의 가능성을 연구하고자 함.
    Ⅱ. 연구개발의 내용 및 범위
    돌연변이형 SOD1에 의한 응집체 형성과 해체 방법을 연구함으로써 ALS에서 응집체의 역할과 발병원인 및 치료제로서의 단서를 확인함. 돌연변이 SOD1과 상호 결합하는 단백질을 인간 신경세포주에 야생형 SOD1 과 돌연변이형 SOD1을 계속적으로 발현하는 Stable cell line 으로 만든 후 면역침강법과 2Dimensional gel electrophoresis 방법을 이용하여 돌연변이형 SOD1과 결합하는 단백질을 발굴함. 또한, 발굴된 단백질과 상호결합하는 SOD1단백질의 특정 절편을 규명하여 경쟁적 저해제로서의 가능성을 확인함.
    Ⅲ. 연구개발결과
    ● Fluorescence Loss In Photobleaching 방법을 사용하여 돌연변이 SOD1의 응집체 형성을 확인.
    ● Fluorescence Loss In Photobleaching 방법을 사용하여 Hsp104에 의해 돌연변이 SOD1의 응집체가 해체됨을 확인.
    ● 돌연변이 SOD1과 상호결합하는 단백질인 Hsp60을 동정하고 Hsp60의 과발현에 의해 돌연변이 SOD1에 의한 세포독성이 줄어듬을 확인.
    ● 돌연변이 SOD1과 아밀로이드 베타의 상호결합을 확인.
    ● SOD1의 N-terminal 16-40 아미노산과 C-terminal 116-140 아미노산의 특정 부분이 여러 단백질과 상호결합함을 확인
    Ⅳ. 연구개발결과의 활용계획
    본 연구 결과를 국제저널에 2편을 게재할 예정임. 본 연구를 통해 ALS의 발병기전 연구에 도움이 될것이며 ALS 치료제 개발에도 기여할 것으로 사료됨.

    Amyotrophic Lateral Sclerosis (ALS) is a neurodegenerative disease that is caused by motor neuronal loss, resulting in movement and respondence deficiency to stimuli. This project focuses on aggregation process and role of aggregates in the pathogenesis of ALS. In addition, proteins interacting with SOD1 are identified and SOD1 domains that interact with the proteins are determined, which will contribute to the development of novel theraputic strategies.
    II. Scope and Contents
    Aggregation mechanism and disaggreation methods have been studied to understand the role of aggregates and get hints of novel therapeutic strategies. Stable cell lines expressing mutant and wild-type SOD1 were constructed and used to identify proteins interacting the SOD1s. SOD1 domains that interact with the proteins were determined.
    III. Results
    ● Aggregation of mutant SOD1 was studied using Fluorescence Loss In Photobleaching.
    ● Aggregates were dissolved by Hsp104, which was confirmed by Fluorescence Loss In Photobleaching
    ● Mutant SOD1 interacted with Hsp60 and overexpression of Hsp60 reduced cell toxicity of mutant SOD1
    ● Mutant SOD1 interacted with amyloid beta
    ● N-terminal 16-40 aa and C-terminal 116-140 aa of SOD1 were found to be important interaction domains with other proteins
    IV. Expected Contribution
    Our results will produce two papers and contribute to the development of novel therapeutic strategies.
