    Bio-conversion of CO2 to methane using anaerobic reactive membrane system

    The increased use of fossil fuels following rapid industrialization since the Industrial Revolution has resulted in the reduction of global forest area through deforestation, and led to global warming. CO2 to the contribution of warming is very large, more than 80%. Therefore, to solve the problem of global warming efficient control and management of carbon dioxide should be prioritized.
    In order to effectively reduce and control CO2, it is necessary to recycle the generated CO2 or convert it to another useful substance. The reason for this is that since carbon dioxide is present in the atmosphere for a long time, suppression of the generation of carbon dioxide only limits the concentration of CO2 in the atmosphere efficiently.
    Methods for recycling CO2 via conversion to bioactive substances can broadly be categorized into chemical and biological conversion methods. This study focused on the methanation of CO2, which is a biological method. The biological conversion requires less energy because it can be conducted at room temperature and pressure. Further, no supplementary material is required for the conversion, as it proceeds via biological reactions. Methanation, which has been utilized for a long period, is the process of producing methane using hydrogen (H2) and microorganisms as the reducing agent and metabolites respectively. The advantages of using methanation include its stability of operation, higher binding ratio compared to other known biological processes (150g CO2/m3-day) and the fact that it can bind CO2 in high concentrations.
    The system set up for this research can be distinguished from those of existing studies by the manner in which it supplies the H2 reducing agents needed to reduce CO2 gases. To circumvent this issue, bubbleless membrane aeration using hollow fiber membranes was utilized in this study. This method is also beneficial in that it allows just enough methanogens to pass through the membrane, thereby reducing wastage of H2 electron donors. Hollow fiber membranes serve as the substrate, allowing methanogens to form a bio film on its surface and grow stably. The methanization of CO2 using the anaerobic reaction membrane was carried out as follows.
    It has been carried out the optimal operating conditions for the methanation of CO2 using anaerobic reactive membrane by the batch reactor. Semi-Continuous Stirred Reactor was operated to determine the amount of inorganic carbon source in the biological process based on the optimum operating conditions at the batch reactor. It derived the test design parameters for the successive reactor. The maximum methanization of CO2 was observed under the condition that pH was 7~8, ORP was–400mV and the ratio of the CO2 and H2 supply was 1:4 and the biological reaction was stable.
    Based on the derived design parameters, three successive reactors of CRL, CRS1, and CRS2 were fabricated and compared and optimized operation conditions for scale-up were derived and a continuous bioreaction process using anaerobic membrane was established. The CRS1 reactor was operated under the acidic conditions of pH 4∼5 and the CRS2 reactor was operated under the neutral condition of pH 7∼8. The amount of methane produced by CRS1 is about 3 times higher than that of CRS1. This is because the amount of methane generated by acetoslastic methanogen using acetic acid, which is continuously maintained as neutral rather than acidic, is increased by hydrogenotrophic methanogen. It is thought that this is the result of adding to methane production.
    The CRL reactor was seeded with methanogens grown in an acidic enrichment culture. The amount of methane generated maintained the average of 500㎖/day, but it is lower than CRS1, which was operated at the same acidic condition. The proportion of methane in the gases released was lower by around 30% compared to reactor CRS1. This implies that the conversion was less effective than in reactor CRS1. The reason for such observations may be the disparity in the microbial communities of the two set-ups and the fact that the CRL set-up was operated with high ORP. Approximately 97% of the methanogen cultured in the enrichment cultures were of the Methanobacterium oryzae family, and the simple make-up of the microbial community could have amplified the effects of inhibiting factors present in the operating conditions or in the reactants.
    To achieve stable a biological process, it is important to investigate the microorganisms responsible for methane production and to determine their optimal growth conditions. The Bacterial 16S rDNA PCR-DGGE technique was employed to identify the microorganisms responsible for methane production. Further, real-time qPCR was carried to measure the population sizes of these microorganisms, and NGS (Next Generation Sequencing) procedures were carried out for the microbial community analysis.
    Analysis of reactor CRS2 via the 16S rDNA PCR-DGGE procedure confirmed the existence of an acetogen named Sporomusa silvacetica, based on the results of DGGE band sequencing. This explains why the methane production level was higher in reactor CRS2 than in reactor CRS1. Furthermore, by analyzing the results of archaeal 16S rDNA PCR-DGGE, it was concluded that methanogens belonging to Methanosarcinaceae and Methanobacterium generation were responsible for methane production in acidic pH ranges, while methanogens from the Methanomicrobiales, Methanospirillum and Methanocorpusculum genera played a vital role in methane production under neutral pH conditions.
    Results of the real-time qPCR showed that Methanobacteriales are mainly involved in hydrogenotrophic methanogenesis, which produces methanol as the final metaboilic product from CO2 and H2. While, Methanomicrobiales were mainly responsible for acetoclastic methanogenesis produces the acetic acid as the metabolite.
    For microbial community analysis via NGS (Next Generation Sequencing), class level and species level analyses were conducted on the archaeal and the bacterial microorganisms. Class level analysis showed most microbes(99.59%) took part in the methanation process. On a species level, numerous species were identified in addition to the predicted Methanobacterium sp. that utilizes H2 gases as reducing agents to convert CO2 to methane. These include; Sporomusa sp., which is involved in the production of acetic acid; Methanomicrobial sp., which produces methane using acetic acid as a substrate; and Anaerolineae sp., Clostridium sp., and Spirorochaceae sp., which generate hydrogen using the organic acids produced. It was deduced that the species excluding Methanobacterium sp. aided the methanation process in directly by increasing the hydrogen and organic acid concentration within the reactor.
    Based on the optimal conditions identified by operating the CSR (Continuous Stirred Reactor) and microbial analysis, a SUCR (Scale- Up Contunuous strirred Reactor) was designed and operated to determine the feasibility of mass conversion of CO2 to methane via biological processes using anaerobic reactive membranes. On the 40th day of the operation, the average methane production reach ed 103.1 l/day. Production of methane was maintained at steady levels afterwards, suggesting that the mass conversion is potentially feasible.
    The technique using microbes that was explored in this study is more applicable than other CO2 recycling techniques in terms of reducing the effective CO2 level in the atmosphere, and it can potentially contribute to both the reduction of CO2 and the production of renewable energy. In the long-term, it is believed that this method could be utilized as a mass CO2 treatment process if ground breaking bioengineering breakthroughs are made, such as finding a microorganism with significantly improved methanation capabilities.

    산업혁명 이후 급속한 산업화에 따른 화석연료 사용의 증가와 산림 훼손으로 인한 산림면적 감소는 지구 대기중에 포함된 온실 가스량의 급속한 증가를 가져왔고, 이는 지구 온난화라는 결과를 가져왔다. 온실가스중 특히 CO2가 온난화의 기여도면에서 차지하는 비중이 80% 이상으로 매우 크다. 따라서 지구 온난화 문제를 해결하기 위해서는 무엇보다. 이산화탄소의 효율적인 제어와 관리가 우선 되어야 한다.
    CO2를 효율적으로 저감·제어하기 위해서는 발생 CO2를 재이용 하거나 유용한 다른 물질로 전환하는 방안이 강구 되어야 한다. 이는 이산화탄소는 발생 후 대기중에 존재하는 기간이 길어 이산화탄소의 발생 억제만으로는 대기중의 CO2의 농도를 효율적으로 저감 하는데 한계가 있기 때문이다.
    CO2를 유용물질로 재이용 하는 방법은 크게 화학적 전환방법과 생물학적 전환 방법으로 나눌 수 있다. 본 논문에서는 생물학적인 방법, 특히 메탄화에 대해 연구하였다. 생물반응은 상온, 상압에서 이루어져, 에너지 소모가 적으며, 생물반응을 이용하므로, 물질전환에 기타 부재료가 필요 없다는 장점이 있다. 특히 메탄화는 오래전부터 사용되어져 온 생물공정으로써 비교적 안정적 운전이 가능하며, 이산화탄소의 고정화율이 타 생물공정에 비해 높다는 장점(150 gCO2/㎥-day)과 고농도의 이산화탄소도 고정이 가능하다는 장점이 있다.
    본 논문의 기존 연구와의 차별성은 중공사막을 이용한 무기포 멤브레인 전달 방식을 사용하여 CO2를 환원하는데 필요한 수소를 CO2와 반응할 수 있게 공급하는 공정 구현에 있다. 이는 효율적인 수소의 공급과 동시에 중공사막이 일종의 담체 역할을 하여, 메탄 생성 미생물들이 안정적으로 자랄 수 있게 하는 부수적 효과도 기대 할 수 있기 때문에 CO2의 메탄 전환률을 크게 향상 시킬 수 있다. 본 논문에서 행해진 실험과 그로 얻은 결과는 다음과 같다.
    회분식 실험을 통하여 혐기성 분리막(Anaerobic reactive membra -ne)을 사용한 CO2의 메탄전환 생물공정의 최적 운전조건을 도출하였으며, 공정중 무기 탄소원의 공급량을 결정하기 위하여 회분식 반응조에서 얻어진 최적운전을 토대로 반연속식 반응조(Semi-Con tinuous Stirred Reactor)를 운전하여 연속 반응조를 제작 하기위한 설계 인자를 도출 하였다.
    이 결과 pH는 7∼8, ORP는 -400mV, CO2와 H2의 공급은 1:4로 공급하는 조건에서 CO2 메탄 전환률의 최대치를 보였으며, 생물학적 반응 또한 안정적 이였다.
    도출된 설계인자를 토대로 CRL, CRS1, CRS2 세개의 연속 반응조를 제작, 비교 실험 하여 Scale-up을 위한 최적 운전조건을 도출 하였고, 혐기성 분리막을 이용한 연속 생물반응 공정을 확립하였다.
    메탄 발생량에 있어서는 산성조건 (pH 4∼5)에서 운전된 CRS1보다 중성조건(pH7∼8)에서 운전된 CRS2가 약 3배 정도 많은 양을 생산하고 있는데 이는 pH가 산성이 아닌 중성으로 유지를 해줌에 따라 지속적으로 발생하는 Acetic acid를 이용하는 Acetoslastic methanogen에 의한 메탄 발생량이 Hydrogenotrophic methanogen에 의한 메탄 생산량에 더해짐으로써 나타난 결과로 판명 되었다.
    CRL 반응기는 산성조건에서 농화 배양된 Methanogen을 반응기에 식종하여 운전하였는데, 평균적으로 500㎖/day 수준의 메탄 발생량을 유지하였지만, 동일한 산성 조건에서 운전이 된 CRS1과 비교를 했을 때 메탄 발생량이 적다고 볼 수 있으며 배출되는 가스 중의 메탄의 비율도 CRS1보다 약 30% 정도 낮으므로 전환률에서도 떨어진다는 것을 확인할 수 있었다. 이는 상대적으로 높은 ORP에서 운전이 된 것과 미생물 군집의 차이 때문이며,. 미생물 분석 결과 농화 배양된 Methanogen은 약 97%가 Methanobacterium oryzae라는 것을 확인했는데 이처럼 단순한 미생물 군집 구성을 가짐으로써 이들이 운전 조건 또는 반응액 내부의 어떤 인자로부터 저해를 받을 경우에 운전 결과 전체가 영향을 받을 우려를 가지고 있다.
    안정된 생물공정 구현을 위해서는 메탄생산에 관여하는 미생물을 규명하고 이들의 최적 생장 조건을 도출 하는 것이 매우 중요하다 이를 위해 미생물종 확인을 위한 Bacterial 16S rDNA PCR-DGGE 와 미생물 개체수를 확인하기 위한 Real-time qPCR 및 다양한 미생물 군집분석(Microbial community)을 위한 NGS(Next Generation Sequencing)를 실시하였다.
    16S rDNA PCR-DGGE 결과, CRS2 반응기에서는 Acetogen으로 여겨지는 Sporomusa silvacetica라는 미생물이 band sequencing 결과로 확인됨으로써 CRS2 반응기 연속운전에서 CRS1보다 메탄 생산량이 많은 것에 대한 결과를 뒷받침해주고 있다.
    Archaeal 16S rDNA PCR-DGGE 결과를 통해 산성 pH에서는 Methanosarcinaceae와 Methanobacterium에 속하는 Methanogen들이, 중성 pH에서는 Methanomicrobiales, Methanospirillum, 그리고 Methanocorpusculum에 속하는 Methanogen들이 주로 역할을 수행하고 있음을 알 수 있었다.
    Real-time qPCR 결과, Methanobacteriales는 Hydrogenotrophic methanogenesis로 대부분 규명 되었으며 이들은 직접적으로 이산화탄소와 수소를 이용하여 최종 대사산물로 메탄을 생산하는 종이다. Methanomicrobiales는 Acetoclastic methanogenesise들이 대부분으로 대사산물로 아세트산을 생산하는 종이다.
    NGS(Next Generation Sequencing)에 의한 미생물 군집분석은 고세균(Archaeal)과 박테리아(Bacterial)에 대해 강(Class level)과 종(Species level)에 대해 하였다. 강(Class level) 분석 결과 대부분(99.59%)이 메탄 생성에 관여하는 미생물이며 종(Species level)분석 결과 당초에 기대하였던 수소를 환원제로 사용하여 이산화탄소를 메탄으로 전환하는 Methanobacterium sp. 이외에도 다양한 미생물군이 확인 되었는데 크게 아세트산 생성에 관여하는 Sporomusa sp.와 Methanomicrobial sp. 와 같이 생성된 아세트산을 기질로 하여 메탄을 생산하는 종류, 또한 생성된 유기산을 기질로 하여 수소를 생산하는 Anaerolineae sp. Clostridium sp. Spirorochaceae sp.이다, 이들은 반응기내에 수소량과, 유기산을 증가시켜, 결과론적으로 메탄생산량 증가에 도움을 주는 것으로 판단된다.
    혐기성 반응 분리막을 이용한 이산화탄소의 생물학적 메탄전환의 대용량화의 가능성을 확인하기 위하여, 연속반응기 운전과 메탄생성 미생물 분석을 통해 도출한 최적 운전 조건을 토대로 Scale-up 반응기인 SUCR을 제작 운전 하였다. 메탄의 발생량은 운전 40일째에 약 103.1l/day 수준의 메탄이 발생했음을 확인하였으며, 그 후 안정되게 지속적으로 메탄을 생산하여, 대용량화의 가능성을 확인하였다.
    본 논문의 연구는 미생물을 이용한 CO2 재이용 기술로써 CO2 저감 부분에 있어 다른 CO2 자원화 기술에 비교하면 CO2의 순수 저감 효율이 높고 저감과 동시에 대사산물로 유용한 자원인 메탄을 생산하기 때문에 CO2 저감 기술인 동시에 환경 친화적인 신재생 에너지의 생산기술로서의 한 부분을 담당할 수 있을 것으로 판단된다. 장기적으로는 생물공학기술의 발달로 CO2에서의 메탄생성 능력이 획기적으로 향상된 미생물이 개발된다면 대용량의 CO2 처리기술로도 활용이 가능할 것으로 판단된다.

    CO2를 유용물질로 재이용 하는 방법은 크게 화학적 전환방법과 생물학적 전환 방법으로 나눌 수 있다. 본 논문에서는 생물학적인 방법, 특히 메탄화에 대해 연구하였다. 생물반응은 상온, 상압에서 이루어져, 에너지 소모가 적으며, 생물반응을 이용하므로, 물질전환에 기타 부재료가 필요 없다는 장점이 있다. 특히 메탄화는 오래전부터 사용되어져 온 생물공정으로써 비교적 안정적 운전이 가능하며, 이산화탄소의 고정화율이 타 생물공정에 비해 높다는 장점(150 gCO2/㎥-day)과 고농도의 이산화탄소도 고정이 가능하다는 장점이 있다.
    본 논문의 기존 연구와의 차별성은 중공사막을 이용한 무기포 멤브레인 전달 방식을 사용하여 CO2를 환원하는데 필요한 수소를 CO2와 반응할 수 있게 공급하는 공정 구현에 있다. 이는 효율적인 수소의 공급과 동시에 중공사막이 일종의 담체 역할을 하여, 메탄 생성 미생물들이 안정적으로 자랄 수 있게 하는 부수적 효과도 기대 할 수 있기 때문에 CO2의 메탄 전환률을 크게 향상 시킬 수 있다. 본 논문에서 행해진 실험과 그로 얻은 결과는 다음과 같다.
    회분식 실험을 통하여 혐기성 분리막(Anaerobic reactive membra -ne)을 사용한 CO2의 메탄전환 생물공정의 최적 운전조건을 도출하였으며, 공정중 무기 탄소원의 공급량을 결정하기 위하여 회분식 반응조에서 얻어진 최적운전을 토대로 반연속식 반응조(Semi-Con tinuous Stirred Reactor)를 운전하여 연속 반응조를 제작 하기위한 설계 인자를 도출 하였다.
    이 결과 pH는 7∼8, ORP는 -400mV, CO2와 H2의 공급은 1:4로 공급하는 조건에서 CO2 메탄 전환률의 최대치를 보였으며, 생물학적 반응 또한 안정적 이였다.
    도출된 설계인자를 토대로 CRL, CRS1, CRS2 세개의 연속 반응조를 제작, 비교 실험 하여 Scale-up을 위한 최적 운전조건을 도출 하였고, 혐기성 분리막을 이용한 연속 생물반응 공정을 확립하였다.
    메탄 발생량에 있어서는 산성조건 (pH 4∼5)에서 운전된 CRS1보다 중성조건(pH7∼8)에서 운전된 CRS2가 약 3배 정도 많은 양을 생산하고 있는데 이는 pH가 산성이 아닌 중성으로 유지를 해줌에 따라 지속적으로 발생하는 Acetic acid를 이용하는 Acetoslastic methanogen에 의한 메탄 발생량이 Hydrogenotrophic methanogen에 의한 메탄 생산량에 더해짐으로써 나타난 결과로 판명 되었다.
    CRL 반응기는 산성조건에서 농화 배양된 Methanogen을 반응기에 식종하여 운전하였는데, 평균적으로 500㎖/day 수준의 메탄 발생량을 유지하였지만, 동일한 산성 조건에서 운전이 된 CRS1과 비교를 했을 때 메탄 발생량이 적다고 볼 수 있으며 배출되는 가스 중의 메탄의 비율도 CRS1보다 약 30% 정도 낮으므로 전환률에서도 떨어진다는 것을 확인할 수 있었다. 이는 상대적으로 높은 ORP에서 운전이 된 것과 미생물 군집의 차이 때문이며,. 미생물 분석 결과 농화 배양된 Methanogen은 약 97%가 Methanobacterium oryzae라는 것을 확인했는데 이처럼 단순한 미생물 군집 구성을 가짐으로써 이들이 운전 조건 또는 반응액 내부의 어떤 인자로부터 저해를 받을 경우에 운전 결과 전체가 영향을 받을 우려를 가지고 있다.
    안정된 생물공정 구현을 위해서는 메탄생산에 관여하는 미생물을 규명하고 이들의 최적 생장 조건을 도출 하는 것이 매우 중요하다 이를 위해 미생물종 확인을 위한 Bacterial 16S rDNA PCR-DGGE 와 미생물 개체수를 확인하기 위한 Real-time qPCR 및 다양한 미생물 군집분석(Microbial community)을 위한 NGS(Next Generation Sequencing)를 실시하였다.
    16S rDNA PCR-DGGE 결과, CRS2 반응기에서는 Acetogen으로 여겨지는 Sporomusa silvacetica라는 미생물이 band sequencing 결과로 확인됨으로써 CRS2 반응기 연속운전에서 CRS1보다 메탄 생산량이 많은 것에 대한 결과를 뒷받침해주고 있다.
    Archaeal 16S rDNA PCR-DGGE 결과를 통해 산성 pH에서는 Methanosarcinaceae와 Methanobacterium에 속하는 Methanogen들이, 중성 pH에서는 Methanomicrobiales, Methanospirillum, 그리고 Methanocorpusculum에 속하는 Methanogen들이 주로 역할을 수행하고 있음을 알 수 있었다.
    Real-time qPCR 결과, Methanobacteriales는 Hydrogenotrophic methanogenesis로 대부분 규명 되었으며 이들은 직접적으로 이산화탄소와 수소를 이용하여 최종 대사산물로 메탄을 생산하는 종이다. Methanomicrobiales는 Acetoclastic methanogenesise들이 대부분으로 대사산물로 아세트산을 생산하는 종이다.
    NGS(Next Generation Sequencing)에 의한 미생물 군집분석은 고세균(Archaeal)과 박테리아(Bacterial)에 대해 강(Class level)과 종(Species level)에 대해 하였다. 강(Class level) 분석 결과 대부분(99.59%)이 메탄 생성에 관여하는 미생물이며 종(Species level)분석 결과 당초에 기대하였던 수소를 환원제로 사용하여 이산화탄소를 메탄으로 전환하는 Methanobacterium sp. 이외에도 다양한 미생물군이 확인 되었는데 크게 아세트산 생성에 관여하는 Sporomusa sp.와 Methanomicrobial sp. 와 같이 생성된 아세트산을 기질로 하여 메탄을 생산하는 종류, 또한 생성된 유기산을 기질로 하여 수소를 생산하는 Anaerolineae sp. Clostridium sp. Spirorochaceae sp.이다, 이들은 반응기내에 수소량과, 유기산을 증가시켜, 결과론적으로 메탄생산량 증가에 도움을 주는 것으로 판단된다.
    혐기성 반응 분리막을 이용한 이산화탄소의 생물학적 메탄전환의 대용량화의 가능성을 확인하기 위하여, 연속반응기 운전과 메탄생성 미생물 분석을 통해 도출한 최적 운전 조건을 토대로 Scale-up 반응기인 SUCR을 제작 운전 하였다. 메탄의 발생량은 운전 40일째에 약 103.1l/day 수준의 메탄이 발생했음을 확인하였으며, 그 후 안정되게 지속적으로 메탄을 생산하여, 대용량화의 가능성을 확인하였다.
    본 논문의 연구는 미생물을 이용한 CO2 재이용 기술로써 CO2 저감 부분에 있어 다른 CO2 자원화 기술에 비교하면 CO2의 순수 저감 효율이 높고 저감과 동시에 대사산물로 유용한 자원인 메탄을 생산하기 때문에 CO2 저감 기술인 동시에 환경 친화적인 신재생 에너지의 생산기술로서의 한 부분을 담당할 수 있을 것으로 판단된다. 장기적으로는 생물공학기술의 발달로 CO2에서의 메탄생성 능력이 획기적으로 향상된 미생물이 개발된다면 대용량의 CO2 처리기술로도 활용이 가능할 것으로 판단된다.
