[Purpose of Study]
Biospecimens collected from the Korea Genome and Epidemiology Study (KoGES) are a finite
resource. The purpose of this study was to analyze additional biomarkers in biospecimens collected
from KoGES in order to promote and maximize the utility of their information
[Study Method]
This study was carried out by conducting clinical tests on long-term stored serum samples kept
frozen at the National Biobank of Korea (NBK). The samples were transported from the NBK to
Green Cross Labs in a frozen state (-60℃ or below) and stored in a deep-freezer (-60 to –90℃)
until the time of analysis. Temperature during storage and transportation was monitored using a
real-time temperature monitoring system. The samples were registered immediately after arrival on a
laboratory information system used exclusively for this study. Sample identification, information, and
volume were verified at each time of arrival. Samples were thawed and spun down immediately
prior to analysis in order to minimize any loss of sample volume. During the analysis, quality control
was assured by measuring materials three times (before, during and after analysis) and by three-step
cross-checking before uploading the analysis results.
[Study Result]
From October 22, 2019 to March 27, 2020 frozen serum samples from 28,000 individuals were
transported from the NBK to Green Cross Labs. Four deliveries were performed over the study
period. As a preliminary evaluation of data quality, analysis of 50 additional long-term stored serum
samples provided by Korea Centers for Disease Control was performed separately. Reference
materials were analyzed before, during, and after the serum sample analysis in order to determine
bias and precision of the measurement method during the study period. Biospecimens were analyzed
within eight hours of thawing. Cystatin-C, Homocysteine, Magnesium, Phosphate were tested for each
serum sample. Test results were uploaded weekly to an integrated disease management system.
[Conclusions]
This study was performed according to standard protocols for sample transport, sample storage
and clinical testing. Data quality was ensured through a quality management system maintained
during the study period. This study provides reliable and valuable information about biomarkers of
biospecimens collected from KoGES, and thus contribute to solutions to many public health issues.
