In the past few years, many studies have focused on the involvement of cytokines in differentiation, proliferation and maturation of T cells within thymus. The int,erleukin 2(IL 2 ) dependent autocrine pathway of growth stimulation might play a key role in the thymic maturation. Here we attempt to provide the information of the effect of interleukin 2(IL 2) on the cytokines production as the cells differentiated to CD8 or CD4 single positive mature T cells in thymus. For this purpose, we set up the method for the detection of intracytoplasmic(IC) cytokines and surface antigens simultaneously and studied the regulation of IL- 2, interleukin 4(IL - 4) and gamma interferon(IFN-7) expression in murine thymocyte by IL- 2 and TPA plus calcium ionophore(Ca.) activation. The results are as follows 1. The staining method using NP 40 as permeabilization solution and formaldehyde as fixatives was adequate way to detect surface CD4, CD8 and IC cytokines simultaneously. 2. Thymic cells were divided to four groups depending on their surface CD4 and CD8 expression'.CD4+ CD8+ double positive cells, CD4 CD8 double negative cells, CD8+ CD4 single positive cells, CD4+CD8 single positive cells. The treatment by IL 2 increased CD8+CD4 cells and TPA+Ca increased DN cells compared to that by media alone. 3. The expression of IC IL 2, IC IL - 4, and IC IFN y in DP, DN, CD8+ CD4 , CD4+CD8 cells was different between the IL - 2 and TPA+Ca stimulation. With IL - 2 treatment, most prominent IC IL - 2, IC IL - 4 and IC IFN y expression was observed in CD8+ CD4 cells. However, there was no specific difference in the expression of IC IL 2, IC IL 4 and IC IFN y between the cell groups by TPA+ Ca. Treatment. In conclusion, these results suggest that IL 2 and TPA+Ca. Have different effects on the surface expression of CD4 and CD8. And the IC cytokines expression in thymocyte seems to be differently regulated depending on the cell activation signals. The CD8+ CD4 cells was the most important source for the cytokines in IL- 2 stimulation.
