Branched-chain amino acids (BCAAs) are closely associated with metabolic disorders; however, their role in the function of brown adipose tissue (BAT) remains unclear. This study aimed to evaluate the effects of dietary BCAA levels on BAT and systemic metabolism in vivo. Mice were fed low or high-BCAA diets under either normal or high-fat diet (HFD)-induced obese conditions, and metabolic parameters were assessed using glucose tolerance test (GTT), insulin tolerance test (ITT), and cold tolerance test (CTT), along with serum biochemical and histological analyses. Under normal conditions, dietary BCAA levels did not significantly affect metabolic outcomes. In contrast, under HFD-induced obesity, mice fed a low-BCAA diet showed improved glucose tolerance and enhanced thermogenic capacity, as evidenced by improved GTT responses and increased temperature maintenance during CTT. These findings indicate that BCAA restriction has minimal effects under normal conditions but improves metabolic function under obesity, potentially through enhanced BAT-mediated thermogenesis. This study highlights the context-dependent role of BCAAs and suggests that dietary BCAA restriction may represent a promising strategy for metabolic disease intervention.
