    본 연구에서는 잘 확립된 우울증 동물 모델을 이용하여 운동의 항 우울 효과의 신경생물학적 메커니즘을 구명하고자 한다. 정상의 마우스에 하루에 2시간씩 14일 동안 (2h x 14d) 의 반복된 신체구금 스트레스를 처치하면 장기간 지속적인 우울 행동변화를 나타낸다. 이 모델에 대해서 스트레스 후 매일 1시간씩 3주간의 운동을 처치하면 여러 행동 검사에서 우울 행동변화들이 항 우울제인 이미프라민을 처치한 경우와 구별되지 않을 정도로 상쇄되었다.
    본 연구를 통해 운동과 우울증 모델에서 공통적으로 활성화된 지역 스크리닝하며, Microarray 와 신경세포 활성을 통한 항우울 지역을 발굴하여, 운동으로 변화한 우울증 조절의 표적 유전자에 대한 기능적 지도를 만들었다. 특히, 뇌의 편도체내 기저핵은 반복적인 스트레스와 운동에 모두 민감하게 반응하는 신경 표적 지역인 것을 보였다. 분자 및 조직학적 분석 방법을 통해 편도체 내에 발현되는 신경 펩타이드들이 반복적인 스트레스 후에 지속적으로 증가되고, 반복적 운동 처치로 인해 감소되었음을 확인하였고, 이는 스트레스와 운동에 의존적으로 변화하는 행동학적 변화를 통해서 재검증 되었다.
    스트레스 후 각 펩타이드 수용체 길항제 처치는 우울증에 치료효과를 보이는 것으로 나타났으며, 뇌의 편도체 내에 정위법을 통해 과발현 혹은 저발현 시킴으로 우울증 행동 결과가 조절되는 결과를 나타냈다. 또한 운동의 항우울 효과 조절의 표적 유전자에 대한 세부기전 및 후생유전학적 조절에 의한 운동의 장기적 항우울 효과 지속의 분자적 기전 규명하였다.

    Depression is a psychiatric illness characterized by multiple symptoms with heterogeneous causes. Physical exercise is generally held to be beneficial in the treatment of weak and moderate depression and reduces future depression. However, the mechanisms underlying the pathophysiology of depression and antidepressant effects of exercise are not clearly understood. In the present study, we investigated whether stress effects and their reversal by exercise occur via a genomic mechanism in the amygdala, a core part of the limbic system important for regulating mood states. Mice treated with repeated stress showed long-lasting depressive behavior, which was counteracted by treatment with three weeks of daily one-hour exercise. Microarray analysis identified a number of genes whose expression in the amygdala was either upregulated or downregulated after repeated stress, and these changes were reversed by exercise.
    Of these genes, the neuropeptides were selected as representative stress-induced and exercise-responded genes in the BLA. Stereotaxic injection of their receptor agonists within the BLA in normal mice produced depression-like behaviors.
