The present communication describes tumor specificity of Korea native phytagglutinin (PHA) Chacomeles sincnsis Koehne (CSK).
In this experiment, the normal human erythrocytes, lcucocytes, lung and urinary bladder cells and tumor cells such as acute and chronic myelocytic leukemia, acute lymphocytie leukemia, lung cancer, urinary bladder cancer, mice leukemic cells and leucoeytes of multiple myeloma patient were suspended in 0.1% EDTA solution and the agglutination test was performed with CSK phytagglutinins which were extracted by various solutions such as saline, ammonium sulfate (AS) and perchloric acid(PA).
The inhibitory effects of the saliva and sugars on agglutination of tumor cells were studied and the following results were obtained.
The normal cells were not agglutinated by CSK saline extracts, while in a few of non leukemic leucocytes were agglutinated by AS and PA extracts.
The tumor cells in this experiment were susceptibly agglutinated by CSK extracts in order of PA, AS and saline extracts. CSK PA extracts induced agglutination of tumor cells were inhibited by D-galactose and L-arabinose but AS and saline extracts were not.
All kinds of CSK extracts such as saline, AS and PA extracts induced agglutinations of tumor cells were not inhibited by human saliva.
The results of this experiment indicate that CSK agglutinability by the various extracts emphasize the importance of examining in a wide variety of cells in attempting to understand the interactions of PHA with a tumor cell surface. It is proved that CSK extracts are more useful than Con A, wheat germ and Arisaema amurense Maximowiez PHA in order to study cellular surface receptor site of tumor cell.
