This experiment was carried out to find the proper way for shoot induction and elongation in bioreactor culture of Rehmannia glutinosa and acclimatization for field cultivation. Multiplication rate of the explants was significantly improved in the induction media with 1.0mg/L thidiazuron alone. Multiple shoots induced in media(MS media with thidiazuron 1.0mg/L) supplemented with paclobutrazol 0.3mg/L had significantly lower vitrification rate. The presence of paclobutrazol during shoot induction decreased the number of shoots and nodes per shoot. Half strength MS media with 1.2% agar and 3% sucrose improved acclimatization and rooting of shoots derived from bioreactor culture. Shoots were effectively acclimatized and rooted well in vessel with membrane filter. Shoots acclimatized in vessel with membrane filter were healthier and had higher ex vitro survival rate than those without membrane filter in plug tray.
