This study was conducted to get basic information on the morphological characteristics of Codonopsis lanceolata (Sieb. & Zucc.) Trautv. pollen. Morphological characteristics were observed using scanning electron microscopy. Distribution of potassium on the pollen surface was investigated using the potassium sensitive fluorescent dye, potassium-binding benzofuran isophthalate (PBFI) and confocal laser microscopy. C. lanceolata pollen is a single medium-sized grain. The pollen viewed at the equatorial axis is oblate or suboblate spheroidal shape. There are very short spinules over the surface forming a particular surface. It has 9 colpates distributed evenly on the surface.
Colpus length covers more than 2/3 of the distance from pole to pole. The edges of each aperture are lined with spinules. The apertures were visible on the immature pollen from the young closed flower. On the mature pollen, the apertures were fully developed and prominent forming a complete 9-colpate shape. Fluorescence of PBFI was observed evenly on the pollen surface except the aperture areas in both immature and mature pollens, indicating that potassium is distributed evenly on the pollen surface except the aperture areas. This finding is different from the previous observations of heavy distribution of potassium on the aperture areas in barley and sesame pollens.
