To evaluate the allergenic antigenicity of buckwheat flour in korean atopic children, we isolated and partially characterized the bubkwheat flour antigens. Three allergenic fractions in crude buckwheat flour extract(BWF- Cr) have been isolated with sephacryl S-Z00 gel filtration. Crude extract and partially purifed fractions of BWF- I, BWF- II and BWF- g elicized positive skin responses in 100%, 100%, 92. 3% and 92. 3% of atopic children sensitive to the commercial crude buckwheat flour extract, respectively. Crude extract contains at least 33 antigenic components as demonstrated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Electroblotting analysis with SDS-PAGE reveals more than 30 allergenic components of molecular weight between 65kd and 12.5kd that are to bind specific IgE. Among these allergens, seven components of molecular weight with 43kd, 41kd, 37kd, 34. 5kd, 32. 5kd, 22kd and 12. 5kd bound commonly the sera of atopic children tested. These are undoubtly the major allergens of buckwheat flour extract in korean atopic children.
