CPTP1 is a nontransmembrane chickenprotein tyrosine phosphatase similar to human PTP1B (HPTP1B). To investigate whether the phosphatase activity of CPTP1 could be phosphoylated by casein kinase II. In vitro phosphorylation and phosphoamino acid analysis showed that both CPTP1 and HPTP1B were phosphorylated on serine and threonine residues by casein kinase II (CKII). Phosphorylation of CPTP1 by CKII resulted in an inhibition of its phosphatase activity in vitro. Endogenous CPTP1-like rat PTP1 was recongnized as a single protein of 51 kDa in rat adipocytes. But CKII did not interact with rat PTP1 in the insulin treated rat adipocytes. These results suggest that rat PTP1 is not implicated directly to CKII-induced signal transduction cascades.
