Objective: In this study, the anti-inflammatory effects of Gardenia Jasminoides were investigated in HMC-1 human mast cell. To identify which mechanism is involved in the anti-inflammatory mechanism, we investigated the inhibitory effect of GJ on PMA and Ionomycin-induced inflammation.
Methods: The effect of GJ was analyzed by RT-PCR, ELISA and Western blotting in PMA and Ionomycin-activated HMC-1, human mast cells.
Results: We found that GJ significantly inhibited the phosphorylation of NF-κB, ERK, JNK, and P38. In addition, GJ decreased nuclear translocation of NF-κB and the mRNA expression of IL-6, IL-8 and IL-17A. GJ also suppreseed the expression of pro-inflammatory cytokines such as IL-6 and TNF-α.
Conclusion: These results suggest that GJ has an anti-inflammatory effect through the phosphorylation of the NF-κB and MAPK proteins, thereby decreasing nuclear translocation of NF-κB and production of pro-inflammatory cytokines in HMC-1 mast cell.
