We demonstrated th use of in vitro amplification of Y chromosome-specific repeated DNA sequences for rapid determination of fatal sex with amniotic fluid, chorionic villus and buccal smear samples. The oligonucleotide primers were synthesized for a 149 bp DNA segment within 3.4kb(DYZ1 family) Y chromosome-specific sequendes. The amplified DNA segments could be observed as bright bands in the expected size range of 8% polyacrylamide gel after 30 amplification cycles in samples of male DNA. No PCR products were observed in samples of female DNA under the same conditions. Polyerase chain reaction can be used as a rapid and useful method for the prenatal sex determination.
