    Six procedures has been presented for the extraction and separation of the total carotenoids in a potent intra-cellular-lipid-producing yeasts. Rhodotorula glutinis var. glutinis sw-17 Two of the methods showed optimum conditions for extraction of the total carotenoid pig ments produced by the above strain and were improved as the routine procedure for the quantitative analysis of yeast carotenoids.
    And the method 2 was more favorable than the method 1. The methods were as fellows
    1.After addition of refined sea sand in yeast suspension, the mixture was dried at 60℃ for 5 hours in oven and grinded with mortar. The Pigments were than extracted from the cells with acetone.
    To ensure the complete extraction of the pigments, 0.5-HCl solution was added to the mixture (least cells and sect sand) separated from acetone and were boiled in water bath at 95℃ for 10 min.
    The pigments extracted from the mixture were boiled with acetone as before.
    These extracts were added together with those obtained from the direct extraction for total caroteroids.
    2.The mixture (1 feast and sea sand) was dried in a rotary evaporator under some vaccume condition at 50℃ for 7 hours.
    And then the pigments were extracted according to the above mentioned method.
