Ⅰ. 연구개발의 목적 및 필요성
연구 필요성
비만은 “ 대유행(epidemic)” 이라 지칭될 정도로 심각하며 이의 증가 추세는 계속될 것으로 전망되고있음. 비만은 단순한 체중 증가 외에, 당뇨병, 고지혈증, 심혈관계 질환, 그리고 일부의 암등 각종 대사증후군의 증가를 수반하여 비만의 연구와 치료제 개발은 더욱 큰 관심사로 여겨지고 있음. 현재 당뇨병의 치료 및 대사 이상을 개선하기 위해 사용되는 약물은 어느정도의 증상을 개선시키나, 체중증가(인슐린, 설폰요소제 및 TZDs), 저혈당 유발(설폰요소제), 부종(TZDs), 및 복통 및 설사(Metformin) 등의 부작용을 가지고 있음. 따라서, 비만과 이의 합병증인 당뇨등의 증상완화 등을 위한 PPARgamma의 활성을 증가시키는 리간드 대신에 새로운 약물과 이를 위한 타겟확보가 시급한 실정임. 새로운 타겟으로 본연구 팀은 Notch 신호 전달체계의 비만과 당뇨에서의 기능 확립과 치료제로의 이용 가능성에 대해 연구를 하고자 함.
연구 목표
Notch 수용체를 통한 신호 전달체계가 비만과 당뇨 발병에서 가지는 역할을 탐색하며, Notch 신호전달 체계를 타겟으로 하는 새로운 비만/당뇨 치료제 개발의 토대를 확립함.
II. 연구개발의 내용 및 범위
Notch는 세포막 수용체로서 발생 과정중에 세포의 분열/사멸과 분화에 관여하며 다양한 조직에서 발현됨. 특히,지방세포 분화와 관련된 보고들에 의하면 Notch의 기능이 비만이나 대사질환과 연관이 있으나,비만과의 상관 관계에 대한 연구가 필요함. 또한, 비만과 연계된 당뇨병의 병태 생리에 중요한 요소인 베타세포의 생존에 Notch의 기능이 보고되고 있으며, 이에 따라 Notch의 당뇨병에서의 역할에 관한 연구도 필요함. 따라서,비만과 당뇨의 연계 연구를 위하여 본 연구팀은 Notch의 역할을 비만과 당뇨병에 초점을 맞추어 연구하고자 하며 내용은 아래와 같음.
1.베타세포주에서 Notch의 활성 정도를 산화스트레스나 지방세포 분화 과정중에 관찰하고, 산화스트레스와 인슐린 저항성,베타세포사멸,지방세포 분화에 대한 효과를 관찰함.
2.당뇨병 질환 모델쥐의 췌장에서 NICD의 세포내 위치 및 하위 유전자들의 발현을 연구함.
3.지방세포의 분화 과정에 Notch 관련 유전자 및 Notch의 억제와 활성에 따른 변화 양상을 연구함.
4.Notch anti-sense mice에서 췌도의 변화와 지방세포 분화 능력을 조사함.
5.Notch의 발현을 억제하는 Notch anti-sense mice의 대사에 대한 분자생물학적,생화학적,조직학적인 연구를 수행함.
6.Notch의 기능을 비만, 당뇨 모델쥐에서 확립하고, Notch의 억제 혹은 활성화를 유도하여 비만과 당뇨병의 타겟으로서의 가능성을 연구함.
III. 연구개발결과
본 과제에서는 Notch의 기능을 중심으로 비만과 당뇨의 연계 연구를 수행하였음. 베타세포에서 Notch의 발현과 활성이 확인되었고, 인슐린 촉진에 관여함을 관찰함. H2O2 투여와 GLP-1 처리는 gammasecretase의 활성 조절로 NICD의 핵내로 이동을 변화시켰음. 이러한 결과들은 Notch와 당뇨와의 밀접한 관계를 보여주고 있음. In vivo 연구에서 Notch의 발현이 감소된 Notch anti-sense mice (NAS)에서 크기가 작은 췌도가 대조군에 비해 증가되었으며, 인슐린 분비의 증가와 GTT에서 대조군보다 우수한 혈당 조절 효과를 나타냄. 또한, Notch의 기능은 지방세포 분화와 고지방식이에 의해 증가됨을 관찰하였으며, Notch의 활성조절은 지방세포 분화를 조절함을 확인함. 고지방식이에 의한 몸무게의 증가는 NAS mice에서 정상쥐와 비교하여 더욱 증가되었으며, Insulin tolerance test에서 인슐린 민감성이 증가됨을 확인함. 이처럼, Notch의 기능은 비만과 당뇨에 직접적인 역할을 할 수 있으며, 본 연구는 비만과 당뇨치료제의 타겟이 될 수 있음을 증명함.
IV. 연구개발결과의 활용계획
본 연구는 비만과 당뇨병의 연계를 통한 발병기작을 규명하고, 이를 통해 독창적이고 효과적인 치료제개발로 발전 시킬 수 있는 계기를 마련 할 수 있음. 당뇨병에서 Notch의 기능조절은 베타세포의 기능을 회복시켜 당뇨병을 완치시킬 수 있는 방법중의한가지가 될 것이며, 향후 줄기세포에서 베타세포로의 변환기전의 이해를 돕고 이를 표적으로 하는약제의 개발에도 도움을 줄 수 있음. 비만에서 Notch의 기능을 파악하여, 지방세포 분화의 이해를 돕고, 새로운 비만 및 대사 관련 치료제의 토대를 마련 할 것임. 본 연구의 성공은 새로운 비만과 당뇨병의 타겟확보로 식품산업,각종 바이오 산업과 제약 산업까지 국제 경쟁력 향상에 기여할 뿐아니라 학문의 활성화에도 기여할 것으로 예상됨.

Significance
Worldwide epidemic obesity is a leading risk factor for insulin resistance, hypertension, hyperlipidemia, and cardiovascular complications, collectively referred to as metabolic diseases. Given the prevalence of obesity and its associated medical problems, new strategies are required to prevent or treat obesity and obesity-related metabolic effects. To this end, we propose to elucidate roles of Notch signaling in obesity and diabetes and further provide therapeutic targets for obesity and diabetes.
Purpose
Our research goal is to identify the roles of Notch signaling in insulin secretion in pancreas and adipogenesis in preadipocytes and further show its beneficial effects on obesity and diabetes.
Ⅱ. Contents
Notch signaling is a conserved pathway that regulates proliferation, differentiation, self renewal potential, apoptosis, and cell-fate decision. Since Notch signaling is expected to play roles in beta cell and adipocytes, we propose to investigate the effects of Notch signaling in these two cell lines. Additionally, we determine the expression profiles of Notch signaling in animal disease models. Finally, we utilize the Notch1 antisense (NAS) mice to investigate the roles of Notch in diabetes and obesity. The included studies are shown below: We investigated the expression profiles of Notch in pancreas under conditions of oxidative stress, insulin resistance and in adipocytes. To define the roles of Notch in pnacreas and adipogenesis, loss and gain of function studies were performed. We also investigated in vivo effects of the Notch signaling in high-fat diet induced obese mice models. We determined the insulin, gluocse levels and body weight of the NAS mice fed with high fat diet.
Ⅲ. Results
We observed the expression of Notch in pancreas under oxidative stress condition. H2O2 treatments changed nuclear localization of NICD in beta cell lines. GLP-1 induced nuclear localization of NICD through inducing gamma secretase activity, suggesting the roles of Notch signaling in diabetes. Total islet number was affected by Notch-1 as shown in notch-1 anti-sense mice. Enhanced insulin and decreased glucagon levels were detected in NAS mice. Accordingly, higher levels of insulin may confer the improved blood glucose tolerance effects in the NAS mice. To also define the function of the Notch pathway in adipogenesis, Notch signaling was genetically and pharmacologically inhibited in preadipocytes. Suppression of Notch1 expression and forced expression of activated Notch “ NICD” affected adipogensis. NAS mice fed with high fat diet decreased lipid accumulation in liver compared to liver from control group and led to the improvement of insulin sensitivity. Taken together, our data elucidate roles of Notch signaling in obesity and diabetes and further suggest the Notch signaling as therapeutic targets for obesity and diabetes.
Ⅳ. Expected contribution
1. Through the successful research, we can potentially identify etiology of obesity and diabetes. Furthermore, this study will offer new insight into therapeutic targets against obesity and its related metabolic diseases such as type 2 diabetes.
2. Mechanical studies for adipogenesis in preadipocytes and insulin secretion in beta cells will help to further understand biology and provide new therapeutic targets.
3. This research also can potentiate the Korea food industry and even pharmaceutical industry with international competitiveness with following activation of related industry and research.
