    트레할로스는 우수한 안정화 능력과 생체적합성을 갖는 비환원성 이당류로서, 식품·화장품·의약품 등 다양한 제형에서 기능성 첨가제로 널리 사용된다. 그러나 현재 산업적 트레할로스 생산은 주로 전분 또는 말토덱스트린을 기질로 하는 maltooligosyl trehalose(MTSase/MTHase) 경로에 의존하고 있으며, 기질 특이성이 좁은 MTSase 특성 및 반응 중 축적되는 가지친 잔기와 저분자 부산물로 인해 정제 공정이 복잡해지고 전체 전환율이 제한되는 구조적 한계를 가진다. 이에 말토스와 트레할로스의 가역적 상호전환을 촉매하는 trehalose synthase(TreS)가 이러한 한계를 완화하고 생성물 조성을 단순화할 수 있는 대안적 생촉매로 주목받고 있다.

    종합하면, 본 연구는 세포 표면에서의 전분 가수분해와 세포 내 TreS 매개 이성화 반응을 결합한 통합 one‑pot 전세포 생촉매 시스템을 제시함으로써, 기존 MTSase/MTHase 기반 트레할로스 공정의 구조적 한계를 완화할 수 있는 기술적으로 실현 가능하고 경제성 잠재력이 높은 대안을 제시하였다. 이러한 플랫폼은 앵커링 모티프, TreS 효소 특성, 공정 운전 조건 등을 추가적으로 개선함으로써 보다 경제적이고 지속 가능한 트레할로스 바이오제조 공정으로 확장될 수 있으며, 향후 다양한 다단계 탄수화물 전환 반응에 적용 가능한 모듈형 one‑pot 전세포 촉매 시스템의 기반을 제공한다.

    Trehalose is a non-reducing disaccharide with excellent stabilizing properties and biocompatibility, which underlies its widespread use as a functional ingredient in food, cosmetic, and pharmaceutical formulations. Nevertheless, most industrial trehalose is still manufactured via the maltooligosyl-trehalose (MTSase/MTHase) pathway using starch or maltodextrin, a process intrinsically constrained by the narrow substrate specificity of MTSase and the concomitant accumulation of branched residues and low-molecular-weight by-products that complicate downstream purification and limit overall conversion. Trehalose synthase (TreS), which catalyzes the reversible interconversion of maltose and trehalose, has therefore attracted attention as a mechanistically distinct biocatalyst that could streamline product spectra and mitigate these limitations when embedded in appropriate process configurations. In this study, Pseudomonas putida KT2440 was engineered as a whole-cell biocatalyst for one-pot trehalose production from starch-based substrates by functionally coupling surface-displayed α-amylase with permeabilized TreS-containing cells. A Bacillus derived from α-amylase was genetically fused to two Pseudomonas outer membrane anchoring motifs, ice nucleation protein (INPC) and OprF, to generate INPC-AmyE and OprF-AmyE surface display systems, and successful presentation on the cell surface was confirmed through extracellular hydrolysis of maltodextrin to maltose. Reaction parameters for these surface-displayed biocatalysts were optimized with respect to pH, temperature, substrate concentration, and whole-cell loading, allowing quantitative comparison of anchoring motifs and assessment of reagent-grade versus food-grade maltodextrin as industrially relevant feedstocks. Kinetic analyses using maltodextrin as the model substrate demonstrated that both display systems supported efficient maltose formation, with INPC-AmyE generally affording higher initial reaction rates and slightly superior catalytic productivity per unit biomass than OprF-AmyE under substrate-saturating conditions. Time-course experiments at varying substrate levels enabled estimation of apparent Michaelis–Menten parameters for the INPC-AmyE whole-cell catalyst and showed that food-grade maltodextrin exhibited a lower apparent 𝐾𝑚 but a modestly reduced 𝑉𝑚𝑎𝑥 compared with reagent-grade material, indicating that low-cost food-grade substrates can be employed without substantial loss of performance. To implement the trehalose-forming step, wild-type P. putida KT2440 cells harboring endogenous TreS were permeabilized with Tween 20 and utilized as whole-cell biocatalysts. The effects of surfactant concentration, pH, temperature, and maltose concentration on trehalose formation were systematically evaluated, identifying near-neutral pH (7.0) and moderate temperature (30 ℃) as optimal and revealing a trade-off between trehalose titer and conversion at elevated substrate concentrations, most likely due to osmotic and membrane stress in permeabilized cells. Under these optimized conditions, the permeabilized cells converted maltose to trehalose while retaining the operational advantages of a self-immobilized enzyme matrix. The two reaction steps were subsequently integrated into a one-pot cascade by co-incubating INPC-AmyE surface-displayed cells with permeabilized TreS cells in a single reactor. Systematic variation of maltodextrin concentration, substrate grade, and the mass ratio of permeabilized cells to INPC-AmyE showed that trehalose productivity is governed primarily by the loading of permeabilized cells and that substantial trehalose titers can be obtained from inexpensive food-grade maltodextrin. Under the best conditions tested, the integrated system produced approximately 4 g/L trehalose with a trehalose conversion of 26.6%. In addition, coupling trehalose synthesis to α‑amylase catalysis increased the apparent overall maltose conversion to about 61%, compared with roughly 40% conversion obtained with the surface‑displayed α‑amylase alone. The enhanced conversion observed in the coupled system is consistent with a substrate-pull mechanism, whereby continuous consumption of maltose by TreS relieves product accumulation around α-amylase and drives maltodextrin hydrolysis beyond the equilibrium attainable in the single-enzyme configuration. Analysis of product distributions as a function of the permeabilized cell:INPC-AmyE ratio further indicates that, under the conditions examined, the maltose-to-trehalose step constitutes the relatively more limiting segment of the cascade, suggesting that targeted improvement of TreS activity or permeabilized-cell performance could yield additional gains in overall conversion. Collectively, this work establishes an integrated one-pot platform that couples cell-surface starch hydrolysis with intracellular TreS-mediated isomerization, providing a technically feasible and potentially more economical alternative to conventional MTSase/MTHase-based trehalose production and offering a versatile framework for future scale-up and process intensification of trehalose biomanufacturing.
