나노산업의 발전에 따라 나노입자의 사용은 급속도로 확장되고 있으며 이에 따라 환경 및 인체에 미치는 위해성도 증가하고 있다. 최근 탄소나노튜브를 비롯하여 금속기반 나노입자 등의 독성을 규명하기 위한 다양한 연구가 보고되고 있으며 급성노출뿐만 아니라 만성노출에 따른 생식발생 및 발암성에 대한 독성연구도 활발히 진행되고 있다. 화학물질과 달리 나노입자는 동일 성분(원소)으로 제조한 경우라도 입자의 크기, 전하, 표면화학 등에 따라 그 독성이 크게 달라질 수 있다. 그러나 이러한 가능성에도 불구하고 아직까지 나노입자에 대한 독성연구의 대부분은 나노입자의 분산을 고려하지 않은 상태에서 시험에 적용되는 경우가 많았다. 따라서 본 연구에서는 각종 나노입자의 분산에 사용되는 초음파 에너지 공급을 표준화한 후 시험대상 나노입자를 분산시킨 상태에서 독성시험에 적용하였다.
독성시험으로는 3D 인체피부모델(EpiDerm™)을 이용한 피부자극성 시험과 A549, THP-1 및 SIRC 세포를 이용한 세포독성시험을 실시하였다. 피부자극성시험은 OECD TG439방법에 준하여 실시하였으며 세포독성시험은 나노렉에서 제시한 표준프로토콜에 준하여 실시하였다. 세포독성기전에 대한 연구로는 DNA 분절시험, ROS생성측정 및 Bax, Bcl-2의 유전자 발현을 평가함으로써 세포사멸과 관련된 측정지표를 평가하였다.
소니케이터의 출력 초음파 에너지 7,056J을 공급하기 위한 조건을 설정한 후 6 mL의 0.05 % BSA 분산 용매에 실리카 나노입자를 2.56 mg/mL 농도로 제조한 후 25% 진폭으로 8분간 초음파 분쇄를 실시하고 DLS로 입도 분포를 측정한 결과 실리카나노입자의 분포는 210 nm < Zave < 270 nm 범위에 존재하는 것을 확인하였다. 이는 분산표준화가 확립되었음을 의미한다. 아울러 실리카, 티타늄, 아연, 세리아, 은 나노입자에 대해 OECD TG 439에 따라 3D 인체피부모델(EpiDerm™)을 이용한 피부자극성 시험을 실시한 결과, 시험에 사용한 나노입자는 비자극성 물질로 판단되었다. A549 세포와 THP-1에 대해 세포독성시험을 실시한 결과, 최고농도인 100 μg/mL의 농도에서 세포독성이 확인되었으며 SIRC세포의 독성시험결과 실리카, 티타늄, 세리아, 은 나노입자와는 달리 아연 나노입자는 상대적으로 강한 독성을 나타내었다. 아연 나노입자는 SIRC세포에서 ROS를 생성시키는 것이 확인되었다. 아울러 DNA가 분절되고 Bax 유전자의 발현이 증가되었으며 Bcl-2 유전자의 발현이 감소되는 것으로 보아 아연나노입자의 세포독성기전은 세포사멸에 의한 것으로 판단되었다.

As the nano-industry has been rapidly developed, the use of nanoparticles is rapidly expanding, and the risks to the environment and the human body are also increasing. Recently, various studies have been reported to investigate the toxicity of carbon nanotubes, metal-based nanoparticles, etc., and studies on the carcinogenicity and reproductive/developmental toxicity due to chronic exposure as well as acute toxicity have been actively conducted. Unlike chemical substances, the toxicity of nanoparticles can be greatly changed according to particle size, surface charge, surface chemistry even if they are made of the same component. Despite this possibility, however, most of the toxicological studies on nanoparticles have been applied to the test without considering the dispersion of nanoparticles. Therefore, this study standardized the supply of ultrasonic energy used for dispersion of each kind of nanoparticles, and then applied the dispersed nanoparticles to the toxicity tests.
Skin irritation test was performed using 3D human skin model and cytotoxicity test using A549, THP-1 and SIRC cells. The skin irritation test was carried out according to the OECD Test Guideline 439 and the cytotoxicity test was carried out according to the standard protocol proposed by NANoREG. The cytotoxic mechanism was evaluated by DNA fragmentation, ROS production and gene expression of Bax and Bcl-2.
After setting the conditions to supply the ultrasonic energy 7,056 J of the sonicator, the silicon nanoparticles which were prepared to a concentration of 2.56 mg/mL in 6 mL of a 0.05% BSA were dispersed by ultrasonication with 25% amplitude for 8 minutes. The distribution of silicon nanoparticles was found to be in the range of 210 nm< Zave <270 nm measured by DLS. This means that the standardization of dispersion has been established in our laboratory. As a result of skin irritation test for silicon, titanium, zinc, ceria and silver nanoparticles, the nanoparticles used in the test were judged to be non-irritancy. Cell viabilities of A549 cells and THP-1 were significantly decreased at the highest concentration of 100 μg/mL. Cytotoxic mechanism of zinc nanoparticles in cultured SIRC cells seems to be the generation of ROS which caused DNA fragmentation. Furthermore, the increased expression of Bax gene and the decreased expression of Bcl-2 suggested APOPTOSIS as the cytotoxic mechanism.
