토양으로부터 중성 pretense를 생산 분비하는 Staphylococcus속균수 LAM 91-4를 분리 선별하였다.
중성 pretense생산의 최적배양조건은 glucose 2%, peptone 0.5%, K_(2)HPO_(4) 0.1%, MgSO_(4) · 7H_(2)O 0.05% 조성의 배지(pH7.0)을 사용하여 30℃에서 5일간 배양했을때 효소생산이 최대에 달하였으며 배양액에 냉각된 acetone을 최종농도가 80%되게 서서히 가하여 12000xg에서 15분간 원심한후 회수된 침전물을 조정제효소로 사용하여 효소학적성질을 조사한 결과는 다음과 같다. 최적 pH와 온도는 각각 pH 7.0과 50℃였으며 효소의 pH안정성은 6.0에서 7.0 까지의 범위에서 비교적 안정성이 있었다.
열안정성은 40℃에서 60분간 처리했을때 거의 안정하였으나 70℃에서는 10분간 처리했을때 약 30% 정도 실활하였다.
Mg^(2+)는 본효소의 활성을 증가시킨반면 Ag^(+), Co^(2+), pb^(2+), Hg^(2+), p-CMB 및 EDTA등은 효소활성을 저해하였다.
효소의 기질특이성조사에서 casein(Hammasten)을 제외하고는 분해력이 거의 없었다.
본균주의 배양적 특성, 형태, 생리 및 생화학적 특성을 조사한 결과 Staphylococcus aureus또는 유연균으로 동정되었다.

A strain of Staphylococcus sp. LAM 91-4 which produces neutral pretense was isolated from soil.
Production conditions and properties of the enzyme were investigated.
Among carbon and nitrogen sources tested, glucose 2% and peptone 0.5% was favorable for the production of the enzyme, respectively. The addition of Mg^(2+) to the basal medium increased the enzyme production.
Initial pH and optimum temperature for the enzyme production was pM 7.0 and 30℃, respectively.
The maximal proteolytic activity was observed after 5 days of cultivation under above conditions in the medium (pH 7.0) composed of glucose 2%, peptone 0.5%, MgSO_(4) · 7H_(2)O 0.05%.
The enzyme was precipitated in the cultured broth by the addition of the cold acetone in the concentration of 80% and the precipitated enzyme was recovered by centrifugation.
The optimum pH was 7.0 and optimum temperature was 50℃.
The enzyme was stable in the pH range from 6.0 to 7.0.
The enzyme was most active at 40℃ and lost 30% of its activity by treatment at 70℃ for 10 min.
The enzyme activity was increased by addition of Mg^(2+) and inhibited by Co^(2+), Hg^(2+), Ag^(2+), Pb^(2+), and p-CMB.
The enzyme was acted well on casein (Hammarsten) but it had poor activity against egg-albumin and gelatin.
Used strain LAM 91-4 was identified as Staphylococcus aureus through the morphological and physiological characteristics.
