A detailed genetic map of the locus oxi3, a large mosaic gene coding for the subunit 1 of cytochrome c oxidase in mitochondrial DNA of Saccharomyces cerevisiae, was constructed. Upon Mn^(++) mutagenesis, a large number of oxi3^- mutants was isolated and mutations were mapped. This mapping shows that about 20% of all the mit^- mutants tested carry large deletions and most of them fall into two classes A and B. Members of the same class are indistinguishable by genetic fine structure mapping and, therefore, are regarded as being the result of repeated, site-specific deletion events.
