Temporal changes in the level of unbound intracellular calcium(Ca) were investigated over 24h in pheochromocytoma cells (PC12) cultured in either serum- or glucose-free conditions both serum deprivation and glucose deprivation led to a dramatic increase in cytoplasmic Ca up to 2h after the onset of deprivation. The Ca level that once reached to a peak decreased to normal within 2h in serum deprivation case while that Ca level elevated by glucose deprivation did not decrease at all for several hours (at least 6h). Intracellular Ca remained elevated over 24h in glucose deprivation. Ryanodine but not nifedipine could prominently black the Ca rises by glucose deprivation, suggesting that a Ca release from intracellular Ca stores may be a major source for raising Ca intracellularly in the lack of energy supply
