Bilateral segmental transection and repair with silastic tube of the main trunk of the sciatic nerves in rats were performed. On one side of each rat, all peripheral branches except the tibial nerve were ligated with silk and divided distal to the ligatures.
At 6 or 12 weeks after operation the number of regenerating axons in each tibial nerve was assessed by morphometric analysis.
1) In experimental group, the number and distribution of regenerating axons were greater and more favorable than control group with statistical significance. : at 6 weeks, 2903 ±275 of axon counts in control, 3674 ±312 in experimental group (P<0.001). at 12 weeks, 4586 ±1148 in control, 7692 ± 1590 in experimental group (P<0.001).
2) The evoked action potential at 8 weeks was more developed in regenerating nerve of the experimental group than that of the control group.
With the above experimentnal results, we conclude that the direction of the regenerated nerve fibers can be reoriented and concentrated to the critical point and so these datas indicate that peripheral ligation is a useful adjunct in nerve rehabilitaiton.
