      Amisulpride, a selective antagonist of D2 andD3 dopamine receptors, is used as an antipsychotic drug. Inthis study, we reported a sensitive LC–MS/MS method fordetermining amisulpride concentrations in rat plasma, anda preclinical pharmacokinetic study in the rat. After asimple protein precipitation with acetonitrile containingmethaqualone as an internal standard, the analytes wereseparated on a reversed-phase column with a mobile phaseof 0.2 % aqueous formic acid and acetonitrile (3:7, v/v).
      The accuracy and precision of the assay were in accordancewith FDA guidance for the validation of bioanalyticalmethods. This analytical method was used successfully tocharacterize the time course of the plasma concentration ofamisulpride following oral administration of a single10 mg/kg dose in rats.
