Phenanthrenes are representative compounds found in the genus Dioscorea and have many bioactivities. The recent study confirmed that phenanthrenes were present in higher amounts in the peel than the flesh of Dioscorea. Dioscorea is not highly storable, so most of them are peeled and then vacuum-packed or dried and then powdered before distribution. In this process, many peels are produced as a byproduct, discarded because there is no use. This study was conducted to develop an optimized extraction method for peels for other industries such as cosmetics. As standard for evaluation of the extraction method, we used three compounds isolated from Dioscorea batatas. A quantitative analysis method using HPLC was developed to quantify the phenanthrenes in the extract, and method validation was performed. Quantitative analysis was performed in 1 to 10 μg/mL concentration of the standard compounds and confirmed acceptable linearity, accuracy, and precision. Four different conditions extracted the peels, and quantitative analysis was performed. As a result, it showed the highest extraction efficiency in steaming pretreatment, high ethanol concentration, and high extraction temperature.
