Purpose
Recently, It is reported that matrix metalloproteinase(MMP) destroy blood brain barrier (BBB) and expression of MMP-9 well correlates with infarct size and neurological deficit in an animal model. Quercetin, a natural flavonoid, antioxidants, is known to inhibit expression of MMP. However, the effect of quercetin on infarct volume and recovery of neuronal injury remain unclear. In the present study, we are going to investigate whether quercetin attenuates brain damage in Sprauge-Dawley(SD) rats with cerebral ischemia, as a model of ischemic stroke.
Method
We made photothrombotic infarct models using cold light after infusion of rose bengal 80 mg/kg into femoral vein of SD rats. We check levels & activities of MMP using a Western blot & zymogram analysis at 2h, 4h, 6h, 12h, 24h, 48h, 72h, 96h after ischemia. Animals were received quercetin intraperitoneally after 1 hour of ischemia and their MMP level & activity were also checked at the same way. The infatct size was estimated by Evans blue dye.
Result
MMP expression increased progressively 2 hours after ischemia, and reached maximum level at 24 hours, then persist 5 days at this level and returned to basal level. With quercetin administration, MMP activity & level at 24h, 48h, 72h, 96h reveled significantly reduced in western blot & zymogram analysis. Reduced activity and level of MMP resulted in decreasing infarct size.
Conclusion
In this study, we conclude that quercetin attenuate BBB disruption, decrease MMP expression, suppresses MMP activity and reduce brain injury.
