Clathrin has been known to be involved in the formation of vesicle coat in trans Golgi network (TGN) and plasma membrane. AP 180, a clathrin assembly protein, has been reported to have an important role in promoting the polymerization of clathrin into a clathrin coated vesicle in neurons.
We had a novel clathrin assembly protein (NCAP) gene that had a high degree of homology (95% amino acid sequence similarity, 86% identity) with AP 180 protein when translated into amino acids. To study the expression pattern of NCAP mRNA during the period of synaptogenesis, in situ hybridization histochemistry was carried out. Like AP 180, NCAP mRNA was highly expressed in the brain between the 1st week and third week after birth which represents the period of synaptogenesis in rat nervous system. However the results also showed that NCAP mRNA was expressed in other tissues. In the lung, the high level of expression was shown from the late embryonic stage till adult. The expression in the kidney was shifted from cortical areas to medulla through the development. And the strong expression in the liver was found at embryonic stage but specific expression was not observed after birth.
From these results, NCAP is suggested to be a new clathrin assembly protein that promotes polymerization of clathrin into a clathrin coated vesicle in various tissues.
