    PCR (Polymerase chain reaction) is a method that amplifies a small sample of specific DNA to a large enough amount for examination. The thermal cycler creates an accurate temperature environment required for amplifing DNA in the PCR process. In this study, a quantitative heat flow analysis was conducted to create an accurate temperature environment and less temperature difference in the 96-well plate by generating uniform heat flow profile in the expansion channel shape of the thermal cycler. The shape of the thermal cycler expansion channel was modeled using the 5th order polynomial equation, commonly used to design the contraction section of a low-speed wind tunnel. Additionally, turbulent heat flow analysis in the 96-well plate thermal cycler was conducted to investigate the temperature difference on the 96-well plate affected by the outer wall shape of the thermal cycler. It was confirmed that the maximum temperature difference of the 96-well plate in the thermal cycler with the suggested expansion wall shape is less than 1K. Based on this study, It is also effective to further reduce the temperature difference by deflecting the air flow by installing a baffle within a range that does not affect the velocity uniformity around the outlet of the thermal cycler.
